Transition-metal catalyzed nitrene transfer is the most desirable strategies to introduce nitrogen functionality into organic molecules, which allows straightforward and shortcut access to many N-heterocycles. Although tremendous progress has been made in the past decade, the formation of cyclic amides has traditionally been hindered by competing isocyanate formation. This dissertation describes how the reaction mechanism guided to provide an innovative solution to the unmet challenge in contemporary catalysis. Based on the mechanistic understanding derived from a seamless collaboration between theory and experiment, we developed customized transition metal catalysts, which enable to insert metal-nitrenoid into diverse functional groups with the exceptional selectivity. The approach was successfully utilized to develop several synthetic methodologies: (i) selective formation of 5-membered amides via C−H amidation, (ii) stereoselective haloamidation of alkynes by ligand participation, (iii) harnessing Ir-nitrenoid as a motif of 1,3-dipole for stereodefined procedure via olefin difunctionalization, (iv) oxidative cyclization based on the intermediacy of carbocation for allyl amide synthesis and its asymmetric variant, (v) [4+2] and [4+1] annulations.
전이 금속을 매개 한 나이트렌을 전이하는 촉매 반응은 유기 분자 내게 질소 작용기를 도입할 수 있는 가장 바람직한 방법의 하나며, 이를 활용하면 다양한 헤테로 고리 화합물을 더 간단하게 합성할 수 있다. 지난 10년간 이 방법의 엄청난 진전이 있었지만, 고리형 아마이드 화합물을 형성하는 일은 예전부터 성공하지 못하고 있다. 이 논문은 반응 메커니즘이 어떻게 현대 촉매 화학에서 다년간 해결하지 못했던 위와 같은 문제의 혁신적인 해답을 주는지에 대해 다룬다. 이론과 실험의 원활한 협업으로 도출된 반응 메커니즘의 이해를 바탕으로, 다양한 작용기에 금속-나이트레노이드를 탁월한 선택성으로 삽입하는 맞춤형 전이 금속 촉매를 고안했다. 이 전략을 활용해 다음과 같은 여러 합성 방법론을 성공적으로 개발했다. (i) 탄소-수소 아미드화를 통한 선택적 락탐 화합물 형성 반응, (ii) 리간드의 참여에 의한 알카인의 입체선택적 할로아미드화 반응, (iii) 이리듐-나이트레노이드를 쌍극자로 활용한 입체특이성의 알케인 이중관능화 반응, (iv) 탄소 양이온을 거친 알릴 아마이드 합성법과 그의 비대칭성 반응의 개발, (v) [4+2] 및 [4+1] 고리화 반응.
