The direct and indirect pathways of the basal ganglia have long been thought to mediate behavioral promotion and inhibition, respectively. However, this classic dichotomous model has been recently challenged. To better understand neural processes underlying rewardbased learning and movement control, I recorded from direct (dSPNs) and indirect (iSPNs) pathway spiny projection neurons in the dorsomedial striatum of D1-Cre and D2-Cre mice performing a probabilistic Pavlovian conditioning task. dSPNs tend to increase activity while iSPNs decrease activity as a function of reward value, suggesting the striatum represents value in the relative activity levels of dSPNs versus iSPNs. Lick offset-related activity increase is largely dSPN selective, suggesting dSPN involvement in suppressing ongoing licking behavior. Rapid responses to negative outcome and previous reward-related responses are more frequent among iSPNs than dSPNs, suggesting stronger contributions of iSPNs to outcome-dependent behavioral adjustment. These findings provide new insights into striatal neural circuit operations.
기저핵의 직접 그리고 간접회로는 오랫동안 동물 활동과 강화의 증가 및 억제를
각각 담당하는 것으로 알려져 있다. 그러나 이러한 고전적인 이분법적인 모델은 최근에 제
시되는 실험적인 근거에 의해 반박되고 있다. 우리는 기저핵의 보상기반 학습과 운동조절과
관련된 신경 처리 과정에 대한 이해를 증진시키기 위해 확률적 고전적 조건화 작업을 수행
하는 D1-cre 그리고 D2-cre 유전자조작 쥐에서 직접 그리고 간접회로에 해당하는 배내측 선
조체 중간가시세포들의 활성을 전기 생리학적으로 측정하였다. 직접회로 중간가시세포는 보
상 가치에 대해 증가하는 활동을 보인 반면, 간접회로 중간가시세포는 감소하는 활동을 보
였고, 이는 선조체가 가치를 직접 그리고 간접회로의 상대적인 활성으로 표상한다는 것을
의미한다. 마지막 혀핥기 이후의 활성 증가는 주로 직접회로와 관련이 있었고, 이는 직접회
로가 진행하는 혀핥기를 억제하는 것을 의미한다. 부정적인 보상에 대한 급격한 증가와 이
전 시도에서의 보상에 대한 정보는 간접회로와 밀접한 관련이 있었고, 이는 보상기반 행동
조절에 간접회로가 더 큰 역할을 한다는 것을 의미한다. 이러한 결과는 선조체의 신경회로
기전에 새로운 시각을 제공한다.
