Neural cells such as neurons and neural stem cells have recently emerged as critical candidates for cell therapy for diverse serious neurodegenerative diseases. However, these still have some limitations including unstable and insufficient cell growth and low differentiation efficiency. Therefore, regulating neural cell behaviors is important issue. In this thesis, we engineer physicochemical surface properties for cellular contact by controlling the functionalities of polymeric surface, dimension of nano-patterns, and mechanical properties and ligands of hydrogel, to regulate the neuronal cell growth and fate commitment of neural stem cells. In addition, we reveal the biomolecular mechanism how the cells process the diverse cues from these materials to their cellular behaviors.
뉴런 및 신경 줄기 세포와 같은 신경계 세포는 최근 다양한 신경 퇴행성 질환에 대한 세포 치료의 중요한 후보로 부상했다. 그러나 이는 여전히 불안정하고 충분하지 못한 세포의 성장 및 낮은 분화 효율을 포함한 몇 가지 한계점들을 가지고 있다. 따라서 신경 세포의 행동을 조절하는 것이 중요한 문제라고 할 수 있다. 본 학위 논문에서는 고분자 표면의 작용기, 나노 패턴의 크기 및 하이드로 겔의 기계적 성질과 리간드를 조절함으로써 세포가 접촉할 수 있는 표면의 물리·화학적 특성을 엔지니어링하여, 신경 세포의 성장과 신경 줄기 세포의 분화를 제어하고자 한다. 또한, 세포가 이러한 세포 행동들에 이르기까지 위 생체 재료 물질들로부터 오는 다양한 신호를 어떻게 처리하는지에 대한 메커니즘을 밝히고자 한다.
