Membrane-associated mucin domain-containing glycosylphosphatidylinositol anchor proteins (MDGAs) bind directly to neuroligin-1 (NL1) and neuroligin-2 (NL2), thereby respectively regulating excitatory and inhibitory synapse development. However, the mechanisms by which MDGAs modulate NL activity to specify development of the two synapse types remain unclear. Here, the crystal structures of human NL2/MDGA1 Ig1-3 complex was determined, revealing their stable 2:2 arrangement with three interaction interfaces. Cell-based assays using structure-guided, site-directed MDGA1 mutants showed that all three contact patches were required for the MDGA’s negative regulation of NL2-mediated synaptogenic activity. Furthermore, MDGA1 competed with neurexins for NL2 via its Ig1 domain. The binding affinities of both MDGA1 and MDGA2 for NL1 and NL2 were similar, consistent with the structural prediction of similar binding interfaces. However, MDGA1 selectively associated with NL2, but not NL1, in vivo. These findings collectively provide structural insights into the mechanism by which MDGAs negatively modulate synapse development governed by NLs/neurexins.
MAM domain-containing glycosylphosphatidylinositol anchor proteins (MDGAs)은 neuroligin-1 (NL1)과 neuroligin-2 (NL2)에 직접 결합하여 흥분성 및 억제성 시냅스 발달을 조절한다. 그러나 MDGA가 어떻게 NL에 결합하여 두 시냅스 발달에 관여하는지에 대한 메커니즘은 아직 밝혀지지 않았다. 본 연구를 통해 규명된 NL2/MDGA1 Ig1-3 복합체의 결정 구조는 세 가지 상호작용부위에 의한 두 단백질의 안정한 2:2 결합을 보여준다. 세 상호작용부위에 대한 MDGA1 돌연변이를 이용한 세포 실험을 통해 NL2에 의한 시냅스형성을 억제하기 위해서 세 결합 부위가 모두 필요하다는 사실을 확인하였다. 또한, MDGA1이 NL2에 결합하기 위해서 Ig1 도메인을 통해 Neurexin (Nrx) 과 경쟁한다는 것을 밝혔다. MDGA1과 MDGA2는 NL1과 NL2 모두에 대해 비슷한 결합 친화도를 나타내며, 이는 단백질의 아미노산 서열과 구조 정보가 보여주는 결합 부위의 유사성과 일치하는 결과이다. 그러나 in vivo 실험 결과는 MDGA1이 NL1이 아닌 NL2에만 선택적으로 결합한다는 것을 보여준다. 본 연구는 NLs/Nrxs의 상호작용에 의한 시냅스 발달이 MDGA에 의해 어떻게 조절되는지에 대한 구조적 통찰력을 제시한다.
