Cyclohept-3-3enones were prepared in four to five steps starting from cyclopentanone silyl enol ether in high overall yields (42-75%). Silyl enol ethers of cyclopentanones were converted to dichlorobicyclo [3.2.0] heptanones (75-88%) via [2+2] cycloaddition with dichloroketene generated from trichloroacetyl chloride. Dechlorination of the dichlorobicyclic ketones was performed by treatment with tributyltin hydride in the presence of AIBN to give bicyclic ketones (89-94%). The ketones were either reduced with sodium borohydride or treated with methylmagnesium bromide to give bicyclic secondary or tertiary alcohols (95-98%). The secondary alcohols were subjected to mesylation ($CH_3SO_2Cl$, $Et_3N$/DCM) and fragmentation (n-$Bu_4NF$/THF) condition to give cyclohpt-3-enones (59-64%). On the other hand the tertiary bicyclic alcohols underwent fragmentation spontaneously during mesylation condition to give the corresponding cyclohept-3-enones almost quatitatively. All the cyclohept-3-enones were converted into the corresponding cyclohept-2-enones quatitatively by treatment with p-TsOH.
Similary, thermal [2+2]cycloaddition of silyl enol ethers with dithisubstituted ketene gave the corresponding cycloadducts. However, when the cycloadducts were treated with $Bu_4NF$, $C_1$-$C_7$ bond cleavage occurred exclusively resulting diketones in almost quantitative yield.
6-Methylbicyclo [5.3.0] dec-10-en-2-one having trans configuration between $C_6$-methyl and $C_7$-proton was synthesized by applying the above methodology for cycloheptenone synthesis. The 5-methyl-1-(trimethyl siloxy) bicyclo [3.2.0]heptanone was treated with Grignard reagent of 2-(2-bromoethyl)-1,3-dioxane, and subsequent fragmentation produced the corresponding cyclohept-3-enone. Stereoselective hydrogenation of cycloheptenone was achieved in the presence of rodium on alumina affording a stereoisomer mixture of cis and trans(15:1). Acid catalyzed cyclization of cycloheptanone derivative was carried out by Heathcock's procedure to give 5-methylbicyclo [5.3.0]dec-10-2-one.
Another application of this methodology was carried out to prepare hydroazulenes starting from hydropentalenones. Silyl enol ethers of hydropentalenones were successfully converted to hydroazulenes in good yield by [2+2] cycloaddition, dechlorination, Grignard, and fragmentation reaction.
Additionally applying the same methodology, (±)-Clavukerin A was synthesized starting from 2-methylcyclopentenyl silyl ether in overall yield 36%. The key elements of this synthesis were the formation of substituted β,$\gamma$ -unsaturated cycloheptenone derivative and highly stereoselective hydrogenation of cycloheptenone derivative by rhodium/alumina. Subsequent acid catalyzed cyclization resulted hydroazulenic skeleton. Shapiro reaction was adapted for transformation of the enone to (±)-Clavukerin A Photooxygenation of (±)-Clavukerin A afforded unstable hydroperoxide, (±)-Clavukerin C, which upon treatment with acid underwent rearragement to give (±)-Clavularin A and B.
시클로 펜타논의 실릴 에놀 에테르로부터 4 또는 5 단계를 거쳐 시클로헵트-3-엔온들을 좋은 수율로 얻을수 있었다. 시클로 펜타논의 실릴에놀 에테르를 디클로로 케텐과 열 [2 + 2] 고리화 첨가반응을 하여 디클로로 두 고리 화합물을 얻었다. 이 디클로로 두고리 화합물을 탈 클로린화 한다음, 수소화 붕소 나트륨으로 환원시키거나 메칠 마그네슘 브로마이드와 반응시켜서 두고리 2차알콜이나 두고리 3차알콜 화합물을 합성하였다. 두고리 2차알콜은 메탄 술포닐 클로라이드와 트리 에틸민에서 반응시킨다음 테트라 부틸 암모니움하에서 고리 절단화 반응을 하여 시클로헵테논을 얻었으며 두고리 3차알콜은 메탄 술포닐화 반응에서 직접고리절단화 반응이 진행 되어 좋은 수율로 시 클로헵텐논 유도체를 제조할 수 있었다.
비슷한 방법으로 실릴 에놀 에테르와 두개의 황이 치환된 케텐과 열[2 + 2]고리화 첨가 반응을 시켜서 상응하는 두고리 화합물들을 제조할 수 있었다. 그런데 두개의 황이 치환된 두고리 화합물은 고리 절단반응에서 위의 예와 다르게 고리의 절단반응이 진행되어 원하는 생산물을 얻을수 없었다.
7 위치의 히드리드와 6 위치의 메칠기가 트란스배치를 갖는 6-메틸비시클로[5.3.0]데스-10-엔-2-온 화합물을 앞의 시클로헵테논 유도체의 합성방법을 이용하여 2-메칠시클로펜타논의 실릴 에놀 에테르로 부터 성공적으로 합성하였다. 이반응 도중 시클로헵테논 유도체를 입체선택적으로 수소화 반응을 알루미나에 흡착된 로디움을 사용하여 성공적으로 수행 했다(시스 : 트란스 = 15 : 1).
또한 시클로 헵테논의 제조방법을 히드로 펜탈레논에 직접 적용하여 히드로 아줄렌 유도체를 성공적으로 합성할 수 있었다.
위의 방법을 이용하여 천연물 (±)-Clavukerin A를 2-메칠시클로펜테닐 실릴 에테르로부터 전체수율이 36% 로 합성하였다. 이 천연물 합성에서 중요단계는 시클로헵테논 유도체를 합성하여 높은 입체 선택적으로 수소화 반응을 한 다음 고리화 반응시키고 Shapiro 반응으로 케톤을 제거하여 고리에 이중 결합을 도입하는 반응이다. 또한 합성한 (±)-Clavukerin A를 광화학 산화반응을 통하여 (±)-Clavukerin C를 합성하고 자리옮김 반응을 하여 천연물 (±)-Clavularin A와 B를 합성하였다.
