Comparing biological networks of different species is widely used to understand the evolutionary principles of various organisms. In particular, there have been a number of studies comparing different species based on their protein interaction network. However, they usually focused on predicting unknown interactions of a particular species based on known interactions of other species, or developing algorithms to identify functionally conserved subnetwork based on network topology. Other studies investigated the topological properties of conserved interactions, however, they focused on the biological properties of pairwise interactions rather than those of evolutionarily conserved core network as a whole. In this study, based on the human and mouse protein interaction networks, we identified the evolutionarily conserved core network with orthologous interactions between them. To find out the evolutionary forces that affect the topology of the core network, we analyzed the relationship between the core network and various experimental data. Additionally, we extended the analysis into other species. Taken together, our analysis can provide a new insight how the biological networks have evolved from its original backbone structure.
본 연구에서는 종간 공통적으로 보존되는 공통 네트워크 구조의 비교를 통하여, 진화상에 서로 다른 종들간에 보존되는 단백질 네트워크의 특성을 분석하였다. 유전자 기능 분석을 통해 여러 종들에서 보존되는 네트워크와 특정 종간에 보존되는 네트워크의 기능의 차이를 설명하였다. 또한 위상학적 관점에서 봤을 때, 종별 네트워크에서 중심부에 위치하는 단백질들의 상호작용이 종간에 보존되는 경향이 높으며, 여러 조직 세포에서 상시적으로 발현되는 항존 단백질들의 상호작용이 진화상 거리가 먼 종들에까지 유의하게 보존되고 있음을 보였다. 이러한 결과는 항존 단백질들간의 상호작용이 네트워크의 중심 뼈대구조로서 진화적으로 다양한 종들간에 보존되는 진화적 통제 요인으로 작용할 수 있음을 보여주며, 기존에 일부 종들에서 이루어진 제한적인 분석 결과들과 유전자 서열 진화속도에 대한 분석을, 진화적으로 거리가 먼 대표 종들에서의 네트워크 진화속도에 대한 분석으로 확장하였다는 점에서 의미가 있다.
