Lectin-like oxidized low-density lipoprotein receptor-1 (OLR-1/LOX-1) is known as a scavenger receptor for oxidized low-density lipoprotein (OxLDL) expressed on all type of cells involved in atherosclerosis and restenosis. OLR-1 expressed in endothelial cells and vascular smooth muscle cells induces the expression of pro-inflammatory molecules. Pro-inflammatory molecules such as cytokines and adhesion molecules that cause monocyte recruitment to endothelial cells and vascular smooth muscle cells (VSMCs) may lead endothelial dysfunction, and proliferation and migration of vascular smooth muscle cells. These inflammatory events play an important role in the pathogenesis of atherosclerosis and restenosis.
Although the involvement of OLR-1 in the inflammatory responses was well-known in both endothelial cells and VSMCs, the signaling pathway evoked by OLR-1 activation and the effect of this signal transduction in VSMCs are still unclear. In this research, to figure out the role of OLR-1 on the inflammatory events in VSMCs, the expression of a major adhesion molecule, ICAM-1 was examined. By the results, the ICAM-1 expression was increased by OxLDL, which lead to induce monocyte adhesion to VSMCs. Then the downstream signaling pathway leading the induced ICAM-1 expression via OxLDL-induced OLR-1 was investigated, and the involvement of NF-κB and NOX4 activation was confirmed in this signal transduction.
From these results, OLR-1 is considered as a key factor mediating the inflammatory events for the progression of atherosclerosis and restenosis by VSMCs. Thus, modulation of the expression of OLR-1 and its downstream signaling pathway can be a clue to regulate the pathogenesis of atherosclerosis and restenosis.
렉틴유사산화저밀도지질단백질수용체-1 (Lectin-like oxidized low-density lipoprotein receptor-1/OLR-1/LOX-1) 는 산화저밀도지질단백질 (oxidized low-density lipoprotein/OxLDL)을 수용하는 스캐빈저수용체 (scavenger receptor)로 죽상동맥경화 (atherosclerosis)와 재협착증 (restenosis)에 관여하는 모든 종류의 세포에 발현되는 것으로 알려져 있다. 내피세포 (endothelial cells)와 혈관민무늬근세포 (vascular smooth muscle cells)에 발현되어 있는 OLR-1은 사이토카인 (cytokines), 세포부착분자 (adhesion molecules)과 같은 염증유도분자의 발현을 유도하고 염증유도분자는 단핵백혈구 (monocyte)가 내피세포와 혈관민무늬근세포에 모여 부착되도록 하여 내피기능장애 (endothelial dysfunction)와 혈관민무늬근세포의 이동과 증식을 야기한다. 이러한 염증반응은 죽상동맥경화와 재협착증의 발병과 진행에 중요한 역할을 한다.
내피세포와 혈관민무늬근세포에서 염증반응에 OLR-1이 관여한다는 것이 잘 알려져 있음에도 불구하고, 혈관민무늬근세포에서 OLR-1이 활성화되었을 때 일어나는 신호전달경로와 이러한 신호전달로 인하여 발생하는 효과에 대해서는 아직 명확히 알려지지 않았다. 그래서 이 연구에서는 혈관민무늬근세포에서 일어나는 염증반응에서의 OLR-1의 역할을 밝히기 위하여 주요 세포부착분자인 ICAM-1의 발현을 검사하였고, 그 결과 OxLDL에 의해 ICAM-1의 발현이 증가하였고 또한 이로 인해 혈관민무늬근에 단핵백혈구가 유의미하게 많이 부착되었다. 그리고 나서 OxLDL에 의해 유도된 OLR-1을 통한 ICAM-1의 발현 증가를 일으키는 하위신호전달경로를 조사하였고, 그 결과 NF-κB와 NOX4의 활성이 그 경로에 포함되어 있다는 것을 확인하였다.
이러한 결과를 통하여, OLR-1은 죽상동맥경화와 재협착증의 발병과 진행에 중요한 염증반응을 조절하는 주요 요소라 결론지을 수 있을 것이며, 그러므로 이를 통해 OLR-1의 발현과 하위신호전달경로를 조절함으로써 죽상동맥경화와 재협착증의 발병 및 진행을 조절할 수 있을 것이라 기대된다.
