Cancer-associated fibroblasts (CAFs) are non-epithelial stromal cells present in the tumor microenvironment (TME) and are known to be involved in the entire stages of cancer and affect cancer progression. CAFs have long been considered as a homogenous population, but recent advances in technologies such as single cell sequencing have revealed that CAFs have heterogeneity, and each subpopulation exhibit distinct phenotypes and functions. The heterogeneity of CAFs affects the patient's prognosis and drug response. Despite the importance of such CAFs heterogeneity, the relationship between certain niche factors and CAFs subtype has not been studied yet. In this study, four different molecular subtypes of fibroblasts are induced through cell culture matrices (collagen and tissue-specific ECM) and gastric cancer cell secretome, and molecular characteristics of each subtype were further analyzed. According to cell embedding matrix, fibroblasts in collagen and tissue-specific ECM acquired myofibroblast-like and inflammatory fibroblast-like features each. By addition of cancer cell secretome, cytokine secretion-related genes were more enhanced in both matrices. These engineered fibroblasts can be used for in vitro tumor models that reflect the fibroblast heterogeneity.
암 관련 섬유아세포는 암 미세환경에 존재하는 간질 세포로 암의 전 단계에 관여하여 암의 진행에 영향을 주는 것으로 알려져 있다. 이러한 암 관련 섬유아세포는 오랫동안 하나의 집단으로서 여겨져 왔으나, 최근 단일세포 시퀀싱과 같은 기술의 발달로 암 관련 섬유아세포에도 이질성이 존재하며, 각각의 유형마다 분자적 형질 및 기능이 다르다는 사실이 밝혀지고 있다. 암 관련 섬유아세포의 이질성은 환자의 예후 및 약물 반응성에 영향을 미치기도 한다. 이러한 암 관련 섬유아세포 이질성의 중요성에도 불구하고, 암 미세환경에 있어서 어떠한 주변 인자가 특정 유형의 섬유아세포를 유도해내는지에 대한 연구가 부족하다. 본 연구에서는 세포 배양 매트릭스와 위암세포 분비체를 통해 네 가지 다른 환경을 조성하였고, 그 환경에 따라 각자 다른 분자 유형의 섬유아세포가 유도되었다. 유도된 섬유아세포의 분자적 특징을 분석함으로써 서로 다른 네 가지 유형의 암 관련 섬유아세포를 정의하였다. 이렇게 유도된 다양한 유형의 암 관련 섬유아세포는 향후 체내의 암 미세환경을 더 잘 반영하는 종양 모델 플랫폼을 만드는데 활용될 수 있을 것이다.
