Riboswitch is a non-coding RNA element that modulates transcription or translation. It attracts much attention because it offers clues about the regulation of RNA polymerases and the development of antibiotic drugs. Pre$Q_1$-I riboswitch regulates transcription of operons related to queuosine biosynthesis. Previous structural studies show that it forms a pseudoknot fold by itself. However, the structure of the pre$Q_1$-I riboswitch bound to RNA polymerase is still unknown. Here, we report its structure determined using single-particle electron cryo-electron microscopy. Our cryo-EM structure reveals that a single-stranded RNA is located on the RNA exit channel and no nucleic acids protrude through the secondary channel. Furthermore, the polymerase in the riboswitch complex shows a swiveling movement related to the elongation complex's pausing mechanism. These results are distinct from those of hairpin pause or the backtracking pause. Our structural study will help a deeper understanding of prokaryotic gene regulation by riboswitches.
리보스위치는 전사 또는 번역을 조절하는 비암호화 RNA 요소이며 항생제 개발의 단서를 제공한다는 점에서 주목을 받고 있다. 특히, pre$Q_1$-I 리보스위치는 queuosine과 관련된 오페론 유전자의 전사를 조절하는 리보스위치이며, 그 압타머 도메인의 구조는 이미 알려진 바가 있다. 그러나 RNA 중합효소에 결합된 pre$Q_1$-I 리보스위치의 전체 구조는 알려져 있지 않기 때문에 초저온 전자 현미경을 사용하여 그 구조를 풀고자 하였다. 리보스위치가 결합된 복합체의 구조를 규명함으로써 비암호화 RNA 요소에 대한 이해를 증진시킬 수 있을 것이다.
