Vascular grafts are widely used to replace damaged or diseased blood vessels in cardiovascular patients. However, synthetic vascular grafts uncovered some issues associated with the nonfunctional endothelial layer, chronic inflammatory response, thrombosis, or endothelial hyperplasia. Recently, tissue-engineered vascular grafts have been developed to compensate for functional limitations. Nevertheless, endothelial detachment from the vascular grafts after implantation remains a significant challenge. This research investigated the behaviors for shear-induced detachment of vascular endothelial cells in a controlled biophysical environment of substrate stiffness or ECM composition using time-lapse imaging.
혈관 이식편은 현재까지 다양한 연구를 통해 그 기능이 개선되어 왔지만, 이식 후 발생하는 내피층의 결핍, 만성 염증 반응, 혈전증 또는 내막 증식과 같은 문제점들이 완전히 해결되지 않고 있다. 이러한 문제점들은 혈관세포의 배양을 통한 혈관 이식편의 내피화와 전단응력을 활용한 사전 처리 등을 통해 부분적으로 개선되었지만, 혈관 이식편의 기능을 더욱 개선하기 위해서는 혈관 내피세포의 전단 유래 박리 거동에 대한 깊은 이해가 필요한 실정이다. 본 학위논문에서는 세포 외 기질의 종류 및 기질의 강성에 따른 혈관 내피세포의 전단 유래 박리 거동을 시간에 따라 분석하고 그 양상에 대해 기술하고자 한다.
