Plant flowers are reproductively important parts of the plant, producing pollinater attraction, defense mechanisms, and seeds. However, the molecular biological mechanisms for the opening of these flowers and the release of fragrance are not well understood. Based on the previous study, using virus-induced gene silencing (VIGS) in wild tobacco, aquaporin TIP and PIP genes, known to be involved in petal expansion, and the ABC transporter genes, which are known to be required for active volatile transport, were silenced and the phenotype was observed. VIGS is a post-transcriptional gene silencing method that employs an RNA interference mechanism. By VIGS method, candidate genes were silenced which are selected by time-course bulk RNA sequencing. In $\textit{N. attenuata}$ flower, g15174, aquaporin related gene downregulated flowers showed less opened petal phenotype. In scent analysis by GC-MS, g06500g13148 and g32098, ABC transporter genes, both displayed a significant difference in Benzyl acetone (BA) and benzyl alcohol emission. However, they did not show any statistical difference among benzoic acid, benzaldehyde, and terpene.
식물의 꽃은 생식적으로 중요한 부분으로 화분 매개, 방어 메커니즘 등이 일어나고 씨를 만든다. 하지만 이러한 꽃의 열림과 향기 방출에 관한 분자생물학적 메커니즘은 많이 밝혀져 있지 않다. 선행연구를 바탕으로 야생 담배에서 바이러스 유도 유전자 침묵을 이용해 꽃잎의 팽창에 관여한다고 알려진 aquaporin TIP, PIP gene들과 향기를 능동적으로 수송하는데 필요하다고 알려진 ABC transporter gene을 침묵시키고 표현형을 관찰하였다. 바이러스 유도 유전자 침묵(VIGS)은 RNA 간섭 메커니즘을 사용하는 전사 후 유전자 침묵 방법이다. 시간 경과에 따른 벌크 RNA 시퀀싱에서 일주기성 리듬이 보이는 것을 후보 유전자들이 선택되었고 그 결과, 야생 담배( $\textit{N. attenuata}$) 꽃에서 aquaporin 관련 유전자 g15174의 발현이 낮아진 꽃은 덜 열린 표현형을 보였다. 또한 GC-MS에 의한 꽃의 향기 분석에서 ABC 수송 유전자인 g06500g13148과 g32098을 각각 침묵시킨 꽃에서는 벤질 아세톤(BA)과 벤질 알코올 방출에서 유의미한 차이를 보였으나 벤조산, 벤즈알데하이드, 테르펜 간에는 통계적인 차이를 보이지 않았다.
