Coacervate produced by phase separation of a polymer solution has high viscosity and adhesive properties, but has poor long-term stability due to the properties of the solution. Electrospinning of coacervate can convert it to a solid with good long-term storage stability, but the high viscosity of coacervate makes the electrospinning process impossible. In this study, a co-solvent system was used to prepare a spinnable coacervate solution, and a bioadhesive membrane was fabricated by electrospinning of the coacervate solution. Based on phenolic tannic acid (TA), hydrogen bonding-dominated coacervates composed of poly(vinyl alcohol) (PVA) or hydroxyethyl cellulose (HEC) were prepared, and their spinnable coacervate solutions were made by lyophilization and dissolution with water-ethanol co-solvent (40~60 % ethanol content). The electrospun membrane is a physicochemically stable solid, but when it comes into contact with water, it is converted back to a coacervate having an adhesive property and exhibits an effective hemostatic effect when applied to the bleeding area.
Three-dimensional (3D) printing, which is often used for the shaping of biomaterials, is known to have a higher probability of local defects in manufactured objects than in conventional casting-based methods, but identification method of defects in 3D printed biomaterials has not been reported. In this study, the uncured position of stereolithography 3D printing was investigated using C. elegans, an animal model system for toxicity evaluation. C. elegans has the ability to 'sense' and 'move' to crawl into safe areas but avoid toxic hazardous areas, providing an immediate indication to identify defect areas and sources of toxicity in 3D printed products.
고분자 용액의 상분리에 의해 생성되는 코아세르베이트는 높은 점도와 접착 특성을 갖고 있지만 용액의 특성 상 장기간 안정성이 취약하다. 코아세르베이트를 전기 방사하면 장기 보관 안정성이 우수한 고체로 변환할 수 있으나 코아세르베이트의 높은 점도로 인해 일반적으로 전기방사를 적용할 수 없다. 본 연구에서는 공용매를 사용하여 방사 가능한 코아세르베이트 용액의 제조와 전기방사를 이용한 생체접착용 멤브레인의 제조를 시도하였다. 페놀성 탄닌산(TA)을 기반으로, 폴리(비닐 알코올)(PVA) 또는 하이드록시에틸 셀룰로오스(HEC)로 구성된 수소 결합이 지배적인 코아세르베이트는 극성 공용매 시스템을 사용하면 전기방사 공정이 가능한 점성의 용액으로 전환된다. 방적된 막은 물리화학적으로 안정한 고체이나, 물에 접촉되어 젖으면 다시 접착 상태를 갖는 코아세르베이트로 전환되어 출혈부분에 적용 시 효과적인 지혈효과를 나타낸다. 생체재료의 형상화에도 이용되는 3차원(3D) 프린팅은 기존 주조 기반 제조 제품보다 제작된 물체가 국부적 결함을 나타낼 확률이 높은 것으로 알려져 있으며, 결함 영역은 독성 성분 방출의 원인이 되는데, 이 위치를 정밀하게 식별하는 방법은 보고되지 않았다. 본 연구에서는 독성 평가를 위한 동물 모델 시스템인 C. elegans를 이용하여 광경화 3차원 프린팅의 미경화 위치를 파악하고자 하였다. C. elegans는 안전한 지역으로 기어가지만 독성이 있는 위험한 지역은 피해가는 '감지' 및 '이동' 능력이 있으며, 즉각적인 징후를 제공하여 3차원(3D) 프린팅 제조물의 결함영역과 독성 근원지를 파악하는데 이용 가능함을 확인하였다.
