Aging is associated with declines in biological functions and various age-associated diseases in living organisms. Aging is affected by multiple factors, including genetic and environmental factors. Although many previous studies have identified various longevity-promoting regimens, one important issue is that longevity does not always increase healthy periods but increases the time of frailty in life. Therefore, an important goal in aging research is to devise proper intervention strategies for increasing this healthy period, while reducing the time of frailty. In this study, I aimed to determine the biological functions of novel genetic factors that regulate healthy aging or immunity using the roundworm Caenorhabditis elegans. First, I identified a PTEN variant that retains longevity without health defects by performing a forward genetic screen using long-lived C. elegans insulin/IGF-1 receptor mutants. Second, by performing a reverse genetic screen, I identified C. elegans long intergenic non-coding RNAs (lincRNAs) that affect the lifespan in wild-type and long-lived C. elegans mutants. Third, I characterized the role of the well-conserved mitochondrial aconitase (ACO-2), a Krebs cycle enzyme that catalyzes the conversion of citrate to isocitrate, in innate immunity of C. elegans. Together, my studies will provide important insights into mechanisms by which novel genetic factors regulate organismal physiology, including aging and immunity. These findings will raise the possibility that PTEN, lincRNA, or mitochondrial aconitase serve as therapeutic targets that promote long and healthy life in humans.
노화는 살아있는 생명체에서 생물학적 기능의 저하와 다양한 노화 관련 질환들을 동반한다. 노화에 영향을 미치는 요인들로는 유전적 및 환경적 요인들 등 다양한 요인들이 있다. 지난 연구들로 장수를 유도하는 식이요법 및 운동요법 등 다양한 장수유도 방법들이 밝혀졌으나, 이렇게 유도된 장수는 항상 건강한 시기만 증가시키는 것이 아니라 병들고 허약한 노쇠기 또한 증가시킬 수 있다는 문제점이 있으며 이것은 앞으로 노화 연구분야에서 해결해야 할 중요한 과제이다. 따라서, 건강한 시기를 늘리면서도 노쇠 시기를 줄일 수 있는 적절한 전략을 수립하는 것이 노화 연구분야의 중요한 목표 중 하나이다. 본 연구에서는 예쁜꼬마선충의 건강한 노화와 면역을 조절하는 새로운 유전적 요인들에 대한 연구를 다루고자 하였다. 첫 번째 연구로, 순 유전학적 스크린을 통해 insulin/IGF-1 신호전달경로에서 장수 돌연변이의 장수를 유지하면서도 건강 결함을 개선하는 PTEN 돌연변이를 발굴했다. 두 번째 연구에서는, 역 유전학적 스크린을 통해 야생형 및 장수 돌연변이 예쁜꼬마선충의 수명을 조절하는 긴 intergenic 비암호화 RNA들을 밝혀냈다. 세 번째 연구에서는, 진화적으로 잘 보존된 구연산회로 효소 중 하나인 미토콘드리아 아코니테이스가 예쁜꼬마선충의 내재면역에 어떠한 역할을 지니는지 밝혔다. 본 연구는 노화와 면역을 포함한 생명체의 생리를 조절하는 새로운 유전적 요인들의 작용 기작을 밝히는데 중요한 통찰을 제공할 것이며, 인간에서의 건강한 수명연장을 위한 치료표적으로 PTEN, 긴 비암호화 RNA 그리고 미토콘드리아 아코니테이스의 가능성을 제시한다.
