Coordinated activities of inter-regional brain networks during the post-learning period are thought to be critical for consolidation of memories. Previous studies have investigated whether the amygdala consolidates a variety of memories through interaction with other brain regions after learning, but relatively little has been known about the neural network mechanism for the consolidation of fear memories. It has been well established that auditory input to the lateral nucleus of amygdala (LA) is essential for acquisition and retrieval of auditory fear memory. Accumulating evidence suggests that fear conditioning induces associative plasticity not only in the amygdala but also in regions outside the amygdala, including various stages of the auditory pathways. Despite these findings, it is unknown whether auditory inputs to the LA are necessary during a post-learning period for consolidation of fear memory. To investigate this question, I optogenetically silenced the inputs from both auditory cortex (AuV/TeA) and thalamus (MGm/PIN) in the lateral amygdala (LA) for 15 min immediately following auditory fear conditioning (FC) and examined its effect on fear memory formation in mice. Optogenetic inhibition of both inputs impaired fear memory formation tested 24 h after FC. This effect was specific to long-term auditory fear memory such that the same inhibition did not affect short-term memory and context-dependent memory. Moreover, long-term memory was intact if both auditory inputs were inhibited at much later time points after FC (3 h or 1 d after FC), indicating that 15 min inhibition itself does not produce any non-specific harmful effect on fear memory retrieval. Selective inhibition of thalamic input was sufficient to impair consolidation of auditory fear memory. In contrast, selective inhibition of cortical input disrupted consolidation of remote fear memory without affecting recent memory. These results reveal a dissociated role of auditory pathways to the LA during early post-learning periods for consolidation of long-term fear memory.
학습 후 뇌 부위 간의 조직화된 신경망 활성은 기억 공고화 과정에 중요하다고 생각되어 왔다. 선행 연구에서 편도체가 학습 후 다른 뇌 부위들과 상호작용을 통해 다양한 기억을 강화하는지에 대해 다뤄왔지만, 공포 기억 공고화의 신경망 기전에 대해서는 상대적으로 잘 알려진 것이 없다. 청각 공포 조건화 학습에서 내측 편도체로의 청각 시상과 피질로부터의 입력 경로는 공포 기억의 습득과 회상에 필수적이라고 밝혀졌다. 또한, 일련의 연구에서 공포 조건화 학습이 내측 편도체 뿐만 아니라 청각 시상과 피질에서의 시냅스 가소성을 유도한다는 것을 발견했다. 이러한 발견에도 불구하고 공포 기억의 공고화를 위해 학습 후 기간 동안 내측 편도체에 대한 청각 입력 경로가 필요한지 여부는 알려져 있지 않았다. 본 연구에서는, 생쥐에서 공포 조건화 학습 직후 15 분 동안 내측 편도체로의 청각 피질과 시상의 입력 경로를 광유전학적 방법을 통해 억제하였을 때 24 시간 후 공포 기억 회상이 손상됨을 확인하였다. 같은 조건의 광유전학적 억제가 단기 공포 기억과 맥락 공포 기억에 영향을 미치지 않는다는 것을 통해, 이러한 효과가 장기 청각 공포 기억 회상에 특이적임을 보였다. 또한, 청각 공포 조건화 학습 3시간 또는 1일 후에 청각 시상과 피질의 입력 경로를 억제한 경우 장기 기억이 손상되지 않음을 통해, 15 분 동안의 억제 자체가 공포 기억에 비 특이적으로 유해한 영향을 일으키지 않음을 확인하였다. 더 나아가 청각 시상 입력 경로의 학습 후 선택적 억제는 청각 공포 기억 공고화를 손상시키기에 충분하였다. 반면, 청각 피질 입력 경로의 선택적 억제는 최근 기억에 영향을 미치지 않고, 오래된 기억만을 특이적으로 손상시켰다. 종합적으로 본 연구는 학습 후 초기 기간 동안 내측 편도체에 대한 청각 시상 및 피질 입력 경로가 장기 공포 기억의 공고화를 위해 분리된 역할을 함을 제시하고 있다.
