The on-line measurement and control of cell concentration of Saccharomyces cerevisiae using a laser turbidimeter was carried out. Using a suspension of harvested cells, effects on the turbidity measurement of operating parameters such as agitation speed, aeration rate, and the concentration of antifoam agent were investigated with the intention of developing a more reliable measurement method than the conventional ones. Correlations between the on-line-measured turbidity and the off-line dry cell weight concentration were made at various operating conditions. In experiment, a lumped turbidity which included both cells' and bubbles' contributions was on-line measured and the cell concentration was estimated by using the correlations.
We used an antifoam agent, Antifoam 289, and the effect of its addition to the culture broth appeared to be negligible in the concentration range tested. The aeration rate in the range of 0 to 0.5 vvm, and the agitation speed between 0 and 400 rpm had no significant influence on the sensor response. The response was, however, significantly affected by a high agitation speed of 500 rpm. The influences of these two operating parameters became negligible as the cell concentration increased. For a batch culture in which the cell concentration was lower than ca. 8 g/L, a correlation a priori constructed at the same operating conditions was used to minimize measurement errors caused by agitation and aeration. The correlation constructed in the absence of bubbles in a beaker was satisfactorily used for the on-line measurement of the cell concentration when it was higher than ca. 8 g/L. During the stationary phase of the cells after the depletion of glucose in batch cultures, considerable deviations were observed between the on-line estimate and the actual cell concentration. It is considered that those deviations were due to morphological changes of the yeast cells. The cell concentration could be satisfactorily estimated on-line in the range of up to 35 g/L in continuous cultures with cell recycle. Two turbidostat experiments carried out at 25 and 35 g/L of cell concentration, respectively, were also successful in terms of on-line measurement and regulation of the cell concentration.
균체 농도의 온라인 측정은 생물반응기의 성공적인 운전과 제어에 매우 중요한 인자이다. 기존의 균체 농도 측정법들 중 배양액의 탁도에 의한 방법은 간단하고 정확한 반면 고농도 배양액의 희석이 요구되어진다.
본 연구에서는 비교적 고농도에서도 희석 과정없이 균체 농도의 온라인 측정이 가능한 laser turbidimeter를 이용하여 효모 농도의 온라인 측정과 제어를 수행하였다. 먼저 수확된 효모를 이용하여 비이커와 발효조에서, 배양액의 탁도 측정에 영향을 미치는 교반 속도, 폭기 속도, 소포제 농도와 같은 각종 조업변수 등의 영향을 알아보고, 보다 정밀한 측정을 위해 특정 조업조건하에서의 배양액의 탁도와 세포건조중량간의 상관식을 마련하였다. 기포는 laser turbidimeter의 응용시 가장 큰 오차 유발 요인으로 간주되어지며, 이의 생성은 위에서 언급한 조업변수 등에 의해 주로 영향을 받는다. 기포로 인한 오차를 최소화하기 위해 기존의 방법과는 달리 기포와 균체의 탁도를 모두 반영한 총괄화 된 탁도값을 사용하였다.
균체 농도 측정시 소포제의 첨가로 인한 측정 오차는 거의 없었다. 교반과 폭기로 인해 발생되는 기포의 영향은 교반 속도 400 rpm이하, 폭기 속도 0.5 vvm이하에서는 무시할 만하였으며, 500 rpm의 강한 교반 하에서는 그 영향이 매우 큼을 알 수 있었다. 그러나 이러한 영향은 균체 농도가 증가함에 따라 감소하였다. 따라서 균체 농도가 상대적으로 낮은 회분식 배양에서는 오차를 최소화하기 위해 동일한 조업조건의 발효조에서 얻어진 상관식을 이용하여 균체 농도를 측정하였으며, 연속 배양시 대략 8 gDC/L 이상의 균체 농도에서는 기포가 없는 상태에서 만들어진 상관식을 큰 오차없이 사용할 수 있었다. 회분식 배양 도중 수행한 온라인 측정은 교반 속도 500 rpm일 때를 제외하면 교반 속도에 관계없이 전반적으로 만족스러운 것이었으나, 양분의 고갈과 함께 균체의 성장이 정지기에 듦에 따라 온라인 측정치와 오프라인으로 측정된 실제 균체 농도간에 상당한 편차가 발견되었는데 이는 균체 몰폴로지의 변화에 기인하는 것으로 생각된다. 균체 재순환 연속 배양을 통한 고농도 배양시 균체 농도 35 gDC/L의 범위까지도 비교적 정확한 측정이 가능했으며, 균체 농도 25 gDC/L와 35 gDC/L에서 수행한 turbidostat 배양 역시 측정과 제어 면에서 성공적이었다.
