Inhibitory peptides against protease were studied from a Korean native leech species, Hirudo nipponia. An elastase inhibitor was purified and characterized by using Sephadex G-75 gel filtration, anion exchange chromatography on DEAE-Sepharose and $C_{18}$ reversed phase HPLC column, followed by acetone precipitation and the complete amino acid sequence of the inhibitor was determined. The purified peptide has a molecular weight of 6,119 Da on MALDI mass spectrum. Inhibitory activity of the peptide was examined on the various kind of proteases. Though the inhibitory peptide has a small effect on chymotrypsin and subtilisin, no inhibitory effect on other proteases such as trypsin, pepsin, and papain was observed. The peptide is stable at the wide range of pH 1 to 11 and is heat stable up to 90℃. The sequence of the peptide was compared with other peptide inhibitors. The amino acid sequence of the peptide shows similarity to subtilisin - chymotrypsin inhibitors from barley and maize and Factor Xa inhibitor, antistasin, from Mexican leech.
한국산 흡혈 거머리 Hirudo nipponia 로 부터 사람의 중성구 elastase를 강하게 억제하는 아미노산 57개를 가진 peptide를 여러 종류의 크로마토그래피법으로 분리하였다. 분자량은 6,122 Da으로 아미노산 서열과 질량 분석기에 의해 확인되었다. 우리가 분리한 peptide가 억제하는 elastase는 serine계 protease에 속하므로 다른 종류의 protease도 억제하는지 조사 하였다. 이 peptide는 trypsin, pepsin, papain은 억제하지 않는 반면, chymotrypsin과 subtilisin을 약 20 - 30 % 억제한다. 이 사실은 우리가 분리한 peptide가 elastase 에만 특이성을 가짐을 보여준다. 57개의 아미노산 잔기중에 10개의 cysteine을 가지고 있고 denaturation과 reduction이 잘 안되는 것으로 보아 disulfide 결합을 하고 있을 가능성이 많다. 이러한 disulfide 결합은 이 peptide를 강산과 강염기 그리고, 고온에서도 그 활성을 잃지 않도록 만드는 것으로 보인다. 아미노산 서열이 밝혀 졌을때 다른 종류의 억제단백질과 서열을 비교하여 보았다. N-말단은 보리와 옥수수에서 분리된 chymotrypsin-subtilisin 억제단백질과 유사한 반면, C-말단은 멕시코 거머리에서 분리한 factor Xa 억제단백질 아주 유사하고 특히, cysteine위치는 완전히 일치한다.
