In tandem with the discovery of bioactive proteins for treating various human diseases, there has been growing interest in making protein carriers using nanoparticles. Nanoparticle-mediated protein delivery methods protect encapsulated proteins in an in vivo delivery process, however, not only the cargo proteins were difficult to translocate to the cytosolic fraction after delivery into target cells but also preparation of purified proteins were complicated. To address these issues, by integrating exosomes with a reversible blue light-mediated protein-protein interaction module, I have developed optogenetic exosomes that are capable of both transient docking and intracellular transfer of functional cargo proteins. This novel protein carrier platform could be capable of stable protein transportation in vivo, leading to a new paradigm for protein drug development.
다양한 사람의 질병 치료를 위한 생 활성 단백질들의 발견에 따라, 생체 내 전달을 위한 나노파티클들을 이용한 단백질 전달체의 제작에 관심이 급증하고 있다. 나노파티클 기반의 단백질 전달 방법은 동봉된 단백질들을 보호하여 안전하게 생체 내에서 전달 할 수 있도록 하였으나, 정제된 단백질을 준비하는 것이 복잡할 뿐만 아니라, 전달하고자 하는 단백질들이 세포질 속으로 전달이 되는 것이 어렵다는 단점들을 가지고 있다. 이러한 문제점들을 해결하기 위해, 본 저자는 가역적으로 푸른 빛에 의해 단백질-단백질 결합이 가능한 모듈을 엑소솜에 결합하여, 일시적으로 기능성 단백질들을 탑재하여 세포 내로 전달이 가능한 광유전학적으로 개조된 엑소솜을 새롭게 개발하였다. 이 획기적인 단백질 전달 플랫폼은 생체 내에서 안정적으로 단백질을 전달할 수 있을 뿐만 아니라, 단백질 약물 개발을 위한 새로운 패러다임으로 작용할 것이다.
