Bioproduction of novel natural products can contribute to sustainable development. Among various approaches of systems biology for producing them, genome-scale models (GEMs) have been found to be effective in predicting the comprehensive pattern of metabolism in organisms. In this thesis, GEMs of Streptomyces strains known for representative sources of secondary metabolites were reconstructed through our modeling program based on their genome data, and the pan-reactome of strains was analyzed. In addition, the utility of GEM was proved by applying it to the overproduction of pikromycin. This thesis is expected to provide helpful information for metabolic engineering to optimize production of secondary metabolites. For future works, modeling frameworks for more complex biological networks than metabolic network should be studied to predict biological processes more accurately.
미생물의 생화학 반응을 기반으로 한 천연물 생산은 지속 가능한 발전에 기여할 수 있다. 이들을 생산하기 위해 다양한 시스템 생물학적인 접근법 중에서 유전체 대사 모델은 세포 내에서 일어나는 전반적인 대사 반응을 예측 가능하게 하여 유용하게 이용되고 있다. 본 연구에서는 이차 대사 산물의 주요 생산원으로 알려진 스트렙토마이시스 균들의 유전체 데이터를 기반으로 그들의 유전체 대사 모델을 구축하기 위한 모델링 프로그램을 개발하였으며, 스트렙토마이시스 집단이 지니고 있는 범반응적 특성을 분석하였다. 또한, 피크로마이신이라는 물질의 과생산 전략 수립에 유전체 대사 모델을 이용함으로써 대사 모델의 가치를 증명하였다. 본 연구를 통하여 이차 대사 산물을 목적으로 하는 대사 공학에 필요한 정보가 제공될 것으로 기대되며, 보다 정확한 생명 현상의 예측을 위해서는 대사 네트워크보다 복잡한 구조의 생물 네트워크에 대한 모델링도 개발되어야 할 것이다.
