Cancer can be interpreted a disease with the broken input-output relationship because of heterogeneous mutations, which trigger uncontrolled proliferation or unmatched output phenotype from the input signaling. Here we found the optimal targets for restoration of the input-output relationship of the colon cancer cell with colon cancer network about 37 colon cancer cell lines. We suggested optimal treatments with the phenotype_score and normal_like_score which was defined, and showed its effectiveness with radar charts and the network graph. These findings supported the previous findings of the cancer treatment and also presented novel insight about the restoration of the input-output relationship of the cancer cell. When we looked at the results of cell lines' response from the treatment, we concluded that a TGFR(OFF) mutation had a critical impact on the broken input-output relationship. And TGFR(OFF) mutation is also a reason for the poor results of drug response of CMS 1&3 stage and MSI compared to CMS 2&4 stage and MSS respectively. And we suggest ‘CyclinD(OFF), ERK(OFF)’ as the general treatment for cancer reversion and proved its effectiveness indirectly with the SW620 and LoVo cell_lines with ERK and CDK4/6 related inhibitors. Therefore we suggest that with respect to the input-output relationship of the cell, receptor mutations like TGFR(OFF), which prevents the TGF_beta signalling, can distort the input-output relationship of cell by inhibiting input signaling. So, in case of cell lines without a TGFR(OFF) mutation, they can be restored to normal cell state by the cell line specific optimal treatments or general treatment like ‘CyclinD(OFF), ERK(OFF)’.
암은 이질적인 돌연변이들에 의하여 입력과 출력간의 관계가 깨어진 병이라고 해석될 수 있고 이에 따라 억제되지 않는 세포의 증식과 입력 신호에 따른 출력의 표현형의 불일치의 현상을 나타낸다. 이 연구를 통해 37개의 대장암 세포 주에 대한 대장암 네트워크 상에서 입력과 출력의 관계의 회복을 위한 최적의 약물을 발굴하였다. 최적의 약물은 정의된 표현형 점수와 정상화 점수에 의해서 제안되었고, 이에 대한 검증은 원형 차트와 네트워크 그래프를 통해 이루어졌다. 이러한 발견은 이전의 암 치료에 대한 기존의 결과와 일치할 뿐만 아니라 암 세포의 입력과 출력의 관계 회복에 대한 새로운 통찰을 제시하였다. 각 대장암 세포 주별 약물에 대한 반응의 결과를 살펴볼 때, TGFR(OFF) 돌연변이가 입력과 출력 관계의 불일치에 매우 중요한 역할을 하는 것으로 결론지을 수 있었다. 또한 TGFR(OFF) 돌연변이가 CMS1&3와 MSI가 CMS 2&4와 MSS에 비해 좋지 않은 약물 반응에 대한 이유가 됨을 알 수 있었다. 또한 암 회복을 위한 일반적인 약물로 ‘CyclinD(OFF), ERK(OFF)’를 제시하면서, 이에 대한 유효성을 SW620과 LoVo에 대한 ERK와 CDK4/6 억제제 약물효과를 통해 간접적으로 증명하였다. 따라서 세포의 입력과 출력의 관계에 대한 관점에서 TGF_beta 신호를 막는 TGFR(OFF)와 같은 수용체 돌연변이는 입력 신호를 차단함으로 세포의 입력과 출력의 관계를 왜곡시킬 수 있다. 결론적으로 TGFR(OFF) 돌연변이가 없는 세포 주의 경우, 세포 주 특이적 최적 약물 또는 ‘CyclinD(OFF), ERK(OFF)’와 같은 일반적인 약물에 의하여 정상 세포의 상태로 회복 될 수 있다.
