Glial cells play an important role for regulating brain homeostasis by forming and eliminating synapses during brain development. However, the underlying mechanisms required for synapse formation and elimination are not well understood. In particular, the proteins that regulate synapse formation and elimination remain unknown. Conversely, the role of PKM2 protein, which is highly expressed in astrocytes, is well known in cancer cells but has not been studied in the brain. In this study, we found that astrocytic PKM2-deficient mice did not change the morphology and viability of astrocytes in the hippocampus. Additionally, although the number of excitatory synapses was not changed, the number of inhibitory synapses was significantly reduced. We also found that astrocytic PKM2-deficient mice showed repetitive behavior, behavioral phenotype associated with autism spectrum disorder (ASD) mouse model. Taken together, these results indicate that PKM2 plays an important role in regulating the number of inhibitory synapses and appropriate brain development and behavior.
별아교세포는 필요한 시냅스를 형성하거나 불필요한 시냅스를 제거하는데 핵심적인 역할을 하고, 이를 통해 뇌의 항상성을 유지한다. 하지만 이러한 시냅스의 형성과 제거가 일어나는 데에 어떠한 메커니즘과 단백질이 관여하는 지에 대해서는 아직 연구가 부족하다. 한편, 뇌세포 중 별아교세포에서 높게 발현되는 PKM2 단백질은 그 역할이 암세포에서는 잘 알려져 있으나, 별아교세포에서는 알려진 바가 매우 적다. 본 연구에서는 PKM2 억제 쥐 모델의 뇌에서 PKM2 단백질의 발현이 대부분 없어진 것을 통해 PKM2의 발현이 쥐 뇌에서 별아교세포 특이적임을 확인하였고, PKM2가 없어도 별아교세포의 형태와 생존에는 영향이 없음을 확인하였다. 또한 PKM2 억제 쥐 모델에서 별아교세포의 수와 모양, 그리고 흥분성 시냅스의 수는 변화가 없으나, 억제성 프리시냅스의 수가 통계적으로 유의미하게 감소한 것을 확인하였다. 시냅스 수의 변화는 쥐의 행동에 변화를 줄 수 있기 때문에 여러 행동 실험을 진행하였고, 그 중 자폐 스펙트럼 장애의 대표적인 증상 중 하나인 반복행동패턴을 보임을 확인하였다. 본 연구 결과를 통해 PKM2는 별아교세포에서 억제성 프리시냅스 수를 조절함으로써 뇌의 정상적인 발달과 행동 발달에 중요한 역할을 한다는 것을 밝혔다.
