Depending on sizes, shapes and the kinds of surface stabilizing ligand, gold nanoparticles shows various physical and chemical properties. Especially, it’s call as ‘ultra-small nanoparticle’ if the diameter of the nanoparticle is less than 3 nm and the physical-, chemical properties of ultra-small nanoparticle is obviously different from that of larger size nanoparticles. The attractive properties (small size, high stability, chemoselectivity and ease of detection under transmission electron microscopy) suggest that usage of ultra-small gold nanoparticle in area of cryo-electron microscopy such as single-particle analysis for biomolecule structure determination and cell tomography is highly feasible. In this thesis, the repeatable synthetic method of ultra-small gold nanoparticle was screened and established. Also, for the usage of the nanoparticle in analysis of biomaterials (proteins or cells) using cryo-electrom microscopy, bio-orthogonal formation of covalent conjugation between ultra-small gold nanoparticle and protein which site-specifically non-canonical amino acid incorporated was studied.
금 나노입자는 그 형태와 크기 그리고 표면을 안정화하는 라이간드의 종류에 따라 여러 물리화학적 성질을 가진다. 특히, 입자의 직경이 3 nm 이하인 경우 초미세 나노입자로 취급되며 그 물리적, 화학적 성질에 있어 큰 변화가 나타난다. 초미세 나노입자의 작은 크기와 높은 안정성, 화학선택성 그리고 투과전자현미경 하의 관찰 용이성은 초저온 전자현미경을 이용한 생체분자의 단분자 구조분석 및 세포단층촬영에 매우 유용하게 사용될 가능성이 있음을 시사한다. 본 학위논문에서는 재현성 있는 초미세 금 나노입나의 합성법을 탐색 및 확립하였다. 또한 투과전자현미경을 이용한 생분자 분석에의 적용을 위해 유전코드 확장을 통해 위치-선택적으로 비천연 아미노산이 도입된 단백질과 초미세 금 나노입자의 생직교화학적 연결 방법을 탐구하였다.
