The hepatic lobule, which is a structural and functional unit of the liver, has the complex microenvironments in which parenchymal hepatocytes and non-parenchymal cells coexist. Hepatocytes exhibit different functional levels depending on their location by the oxygen gradient and the transcriptional changes of genes, which is called liver zonation. The engineered liver tissues have reproduced the complex environments for physiologically-relevant in vitro drug testing, but there was limited in analyzing them by location. Here, we present a novel three-dimensional (3D) tissue-level hepatic cell culture platform layering the manipulable collagen sheets in order to analyze the reconstructed liver zonation spatially. The developed micropatterned collagen sheet combined with paper support enabled the manipulation of an extracellular matrix-based sheet module and provided the 3D microenvironments for various cell types. The assembly of the sheets containing hepatocytes and endothelial cells, respectively, created a 3D co-culture environment, which improved hepatic function, and the layer arrangement of the stacked sheets also affected the function. In addition, the sheet micropatterning was able to control the accessibility of oxygen and nutrients in the stacked sheets. The disassembly of the stacked sheets enabled layer-by-layer analysis and allowed to confirm the hepatic zonation qualitatively. A demonstration of acetaminophen-induced liver injury using the stacked sheets showed the improved drug sensitivity with co-culture and chemical drug-metabolic induction and presented the quantitative results of the different cellular responses to the drug by layer according to liver zonation. Therefore, this platform is expected to be used for in-depth drug toxicity assays of complex liver tissue via spatial analysis.
간의 구조적 및 기능적 단위인 간소엽은 다양한 세포가 공존하며 산소 구배 및 유전자 발현 차이에 의해 기능적으로 구역화된 복잡한 환경을 갖고 있다. 기존에 제작된 인공 간 조직은 생리학적으로 유사한 체외 약물 검사를 위해 복잡한 환경을 재현했지만, 위치 별로 분석하기에 한계가 있었다. 따라서, 본 논문에서는 조작 가능한 콜라겐 시트 기반 공간적으로 분석이 가능한 새로운 조직 수준 삼차원 간 세포 배양 플랫폼을 제시한다. 종이 지지대와 결합된 미세패터닝된 콜라겐 시트는 세포외기질 시트 모듈의 조작을 가능하게 하고 다양한 세포 유형에 대한 삼차원 미세 환경을 제공했다. 이를 이용한 시트의 조립은 삼차원 공배양 및 적층 배열에 따른 간 기능의 개선을 가능하게 했고, 시트의 미세 구멍 크기를 조절하여 간 구역화을 위해 적층된 시트 내 산소 및 영양소의 접근성을 제어할 수 있었다. 또한, 적층된 시트의 분해는 층별 분석을 가능하게 하고 간 구역화를 정성적으로 확인할 수 있게 했다. 마지막으로 적층된 시트를 아세트아미노펜에 의해 유도된 간 손상 모델로 사용하여 공배양 및 화학적 대사 유도에 의한 약물 민감도 개선의 가능성을 보여주었고, 간 구역화에 따른 약물에 대한 상이한 층별 세포 반응의 정량적 결과를 제시했다. 따라서, 본 기술은 심층적 약물 유래 간 독성 분석을 가능하게 할 것으로 기대된다.
