It is essential to figure out the structure of the molecule for understanding the properties of molecules and related chemical reactions. As the chemical reaction proceeds, the three-dimensional structure of the molecule changes through metastable states on the reaction coordinates, called reaction intermediates. The structure of the reaction intermediate plays a key role in determining the pathway and product of the reaction, so investigating the molecular structures of the reaction intermediates is fundamental to understand and control the reaction. Time-resolved x-ray solution scattering (TRXSS), also known as time-resolved x-ray liquidography (TRXL), is a relevant tool for probing the ultrafast structural dynamics with high spatio-temporal resolution. Based on the pump-probe method, an optical laser pulse (pump) initiates a reaction and a time-delayed hard x-ray pulse (probe) tracks the progress of the reaction as the scattering pattern. The scattering pattern is sensitive to the global structure of the intermediate as the scattering signal arises from all atomic pairs in the molecule and thus TRXSS can be a complementary tool to the time-resolved spectroscopy tools that are sensitive to specific energy states or chromophores. The time resolution of TRXSS can be achieved by about 100 ps with the 3rd-generation synchrotron radiation source. We performed TRXSS at synchrotron on the I114F mutant of HbI and compared the results of the I114F mutant with those of the wild type to reveal the relationship between ligand migration and the structural changes. With the advent of x-ray free electron lasers (XFEL), the time resolution of TRXSS can be improved to sub-picoseconds. With the higher time resolution aided by XFEL, we observed the in-cage isomer formation mechanism of the CH2I2 and successfully determined the structure of the transiently formed precursor of the isomer.
분자 구조는 분자의 특성 그리고 관련된 화학반응을 이해하는데 있어 근간되는 정보이다. 화학 반응이 일어날 때, 분자는 구조 변화를 겪게 되는데, 이 과정에서 형성되는 준안정 상태를 반응 중간체라 하며, 반응 중간체의 구조는 반응의 경로와 생성물을 결정하는데 중요한 역할을 한다. 따라서, 반응 중간체의 구조를 밝히는 것은 해당 화학 반응을 이해하고 제어하는데 필수적이다. 용액상 시간분해 엑스선 산란법은 높은 시공간 분해능으로 반응 중간체의 구조와 동역학, 즉 반응의 구조 동역학을 관찰하는데 유용한 실험방법이다. 펌프-프로브 방법을 기반으로, 광학 레이저 펄스(펌프)가 반응을 개시하면 일정 시간 뒤에 들어오는 엑스선 펄스(프로브)가 산란 패턴의 형태로 반응의 진행을 추적한다. 산란 패턴은 분자의 모든 원자 쌍으로부터 발생한 신호이기 때문에 분자의 전체구조에 민감하다. 따라서, 용액상 시간분해 엑스선 산란법으로부터 얻어지는 정보는 특정 에너지 상태나 발색단에 민감한 신호를 얻는 시간분해 분광법과 상보적이다. 시간 분해능의 경우, 3세대 싱크로트론 방사원을 사용했을 경우 약 100 ps 의 시간 분해능을 가질 수 있다. 싱크로트론에서의 동종이합체 헤모글로빈의 I114F 돌연변이체에 대한 시간분해 엑스선 산란 연구를 통해, 돌연변이체와 야생형의 결과를 비교하여 리간드 이동과 단백질의 구조변화 사이의 관계에 대해 밝힐 수 있었다. 자유 전자 레이저의 발전으로 펨토 초 영역에서 일어나는 반응을 시간 분해 엑스선 산란법으로 관찰할 수 있게 되었다. 더 높은 시간 분해능의 도움으로 CH2I2 의 케이지 내부에서 일어나는 이성질체 형성 메커니즘을 관찰하며 이성질체의 전구체를 구조적으로 밝힐 수 있었다.
