Drug resistance is a major cause of cancer recurrence and metastasis. Among various cancers, in breast cancer, where metastasis and recurrence occur at high frequency, efforts are being made to overcome chemoresistance with a polychemotherapy using multiple anticancer drugs at the same time, but the key mechanism that causes drug resistance has not been investigated. Metabolic reprogramming is one of the causes of drug resistance by favoring the differentiation and survival of cancer cells by using a different metabolic pathway from normal cells. To study metabolic reprogramming occurring in paclitaxel- and adriamycin-resistant breast cancer cells, we tried to understand the metabolic network through model simulation and multi-omics analysis, and we tried to identify key reactions and genes that influence drug resistance.
약물 내성은 암의 재발과 전이에 영향을 미치는 주요 원인이다. 다양한 암 중에서도 전이와 재발이 높은 빈도로 일어나는 유방암에서 여러 항암제를 동시에 사용하는 복합화학요법으로 화학 저항을 극복하기 위해 노력하고 있지만 여전히 약물 내성의 원인이 되는 핵심 기작을 규명하지 못한 상황이다. 대사 재 프로그래밍은 정상 세포와는 다른 대사 경로를 이용하여 암세포의 분열과 생존을 유리하게 만들어 약물 내성을 일으키는 원인 중 하나로 꼽힌다. 우리는 파클리탁셀과 아드리아마이신 내성 유방암 세포에서 발생하는 대사 재 프로그래밍을 연구하기 위해 모델 시뮬레이션과 다중 오믹스 분석으로 대사 네트워크를 이해하고자 하였고 이를 통해 약 내성에 영향을 미치는 핵심 반응 및 유전자를 밝혀내고자 하였다.
