Aberrant cancer metabolism has been recognized to be an attractive target for cancer treatments. However, inhibition of a single metabolic pathway has limited efficacy. In this study, a strategy to concurrently inhibit tumor glycolysis and oxidative phosphorylation is proposed. An antiglycolytic drug and artificially synthesized mitochondria-targeting polypeptide were simultaneously applied to completely deplete the energy source of cancer cells, eventually leading to a synergetic effect of cancer starvation therapy. $\textit{In vitro}$ results show the mechanism and evidence of the cooperative effect of this combinatorial approach. This study is expected to provide new perspectives of cancer starvation treatments by inhibiting various metabolic pathways.
암세포의 비정상적인 물질대사는 최근 들어 암 치료 표적으로써 떠오르고 있다. 하지만 단일 대사 경로의 억제는 제한된 효과를 가지고 있다. 본 연구에서는 암세포의 해당과정과 산화적 인산화를 동시에 억제하는 전략을 다루고자 한다. 해당과정을 억제하는 약물과 함께 인공적으로 합성된 미토콘드리아 표적형 폴리 펩타이드를 적용시킴으로써, 암세포의 에너지원을 완전히 고갈시켜 대사항암제에 시너지 효과를 가져오게 된다. 암세포에 실험을 진행한 결과를 통하여 이러한 접근방식의 작용 경로 및 협력 효과에 대한 증거를 보여준다. 이번 연구를 통하여 다양한 대사 경로를 억제할 수 있는 대사항암제에 대한 새로운 시각을 제공할 것으로 기대된다.
