Polyelectrolyte complex micelles (PCMs) are nanoparticles formed by the complexation between a charged-neutral hydrophilic block copolymer and an oppositely charged polyelectrolyte. Herein, the effect of N/P ratio and DNA hybridization on morphology of PCMs formed from DNA-poly(ethylene glycol) (DNA-PEG) block copolymer and poly(L-lysine) (PLys) are investigated. Single-stranded (ss-) and double-stranded (ds-) DNA-PEG diblock copolymers are synthesized by solution-phase coupling. PCMs are prepared using thermal- and salt-annealing protocol under different N/P ratio conditions. Direct TEM imaging is used to observe the morphology of the PCMs. The TEM images show that the micelles are spherical in the majority of cases. Rod-like micelles are observed for salt-annealed ssPCMs at N/P=2, and dsPCMs at N/P=2, and 3 are shown to have ellipsoidal shapes. The present study can provide a basis for design and optimization of DNA block copolymer-based PCM systems for further application such as drug encapsulation or gene delivery.
고분자 이온 복합체 마이셀 (Polyelectrolyte complex micelle, PCM)은 친수성 하전-중성 블록 공중합체와 반대 전하를 띠는 고분자 이온 간의 정전기적 상호 작용에 의해 형성 되는 나노 입자이다. 본 논문에서는 N/P비와 DNA 혼성화 상태가 DNA-폴리에틸렌글리콜 (DNA-b-PEG) 블록 공중합체와 폴리라이신 (poly(L-lysine), PLys)으로 이루어진 복합체 마이셀 모폴로지에 미치는 영향을 연구하였다. 단일 가닥DNA와 이중 가닥 DNA를 포함하는 DNA-PEG 블록 공중합체를 완충용액 상에서 커플링 반응을 통해 각각 합성하였고 고온 어닐링 및 염 어닐링을 통해 PLys와 복합화를 진행하여 마이셀을 형성하였다. 투과 전자 현미경법 (transmission electron microscopy, TEM)을 통해 마이셀 모폴로지를 분석하였다. 대부분의 경우 구형 마이셀이 관찰되나 염 어닐링으로 형성된 단일 가닥 DNA-PEG 복합체 마이셀의 경우 N/P비가 2일 때 막대형을 띠며, N/P비가 2이상인 조건 하에서 이중 가닥 DNA-PEG 복합체 마이셀이 타원형을 띰을 확인하였다.
