Spheroid, a 3D aggregate of tumor cells in a spherical shape, has overcome the limitations of conventional 3D cell models to accurately mimic the in-vivo environment of a human body. The spheroids are often cultured with other primary cells and embedded in collagen drops to construct a spheroid–hydrogel model that better mimics the cell–cell and cell–extracellular matrix (ECM) interactions. However, the conventionally used methods of culturing and embedding spheroids into ECM has several shortcomings. The procedure of transferring a single spheroid at a time by manual pipetting results in the inconsistent arrangement and even loss or damage of the spheroid. Based on the previously introduced droplet contact-based spheroid transfer technique, we present a poly(dimethylsiloxane) and resin-based drop array chip and pillar array chip with alignment stoppers, which enhances the alignment between the chips for uniform placement of spheroids and eliminates the need to use a stereomicroscope while handling the cell models. The novel platform allows a homogeneous and time-efficient construction and diverse analysis of an array of fibroblast-associated glioblastoma multiforme spheroids embedded in collagen.
종양세포를 구 모양의 3D 형태로 배양한 스페로이드는 기존 2D 모델의 한계를 극복하고 생체 내 환경을 더 정확히 모사한다. 스페로이드는 세포와 세포, 또는 세포와 세포외기질 간의 상호작용의 정확한 모사를 위해 종종 다른 일차세포와 배양되거나 콜라겐 등의 하이드로겔에 삽입하여 스페로이드-하이드로겔 모델을 만든다. 하지만, 스페로이드를 배양 또는 이식하는 기존의 과정은 여전히 수동 피펫팅으로 일일이 진행되고 있어 스페로이드 모델의 배열이 비일관적이거나 세포 모델의 손상 또는 손실이 발생할 위험이 높다. 따라서, 앞서 발표된 액적 접촉 기반의 스페로이드 이식 기술을 기반으로, 본 연구에서는 정렬 플랫폼이 추가된 새로운 액적 어레이 칩과 기둥 어레이 칩을 제시한다. 이를 통해 섬유아세포가 포함된 종양 스페로이드-하이드로겔 모델을 균일하게 배양 및 이식하고 복잡하고 시간 소요가 많은 현미경의 사용 필요성을 없애 효율적으로 3차원 종양 세포 모델의 다양한 분석이 가능하다.
