Understanding brain circuits is important to develop treatments for brain disorders or to find mechanisms of brain diseases and in vivo and in vitro experiments are conducted to measure neural activity of stimulated specimens. Due to in vitro disease models showed lack of complexity than human brains, brain organoids were developed to mimic complex structure and various functions of the brains. However, current neural recording devices are not suitable to the brain organoids due to its invasiveness and limited recording areas. In this dissertation paper, a novel microelectrode array (MEA) to read biological signals of the brain organoids over a large area was proposed. Serpentine shape patterned flexible and transparent polyimide increased stretchability and bilayer structure have contact with both base and side area of the organoids. Ex vivo mouse brain was placed on top of the MEA and neural signals were recorded successfully, but there is a limitation that could not be measured in the presence of media due to poor contact between the electrodes and the brain.
뇌 질환을 위한 치료법을 개발하거나 뇌 질환의 근본적인 원리를 밝혀내기 위해서는 뇌 회로를 이해하는 것이 중요하며 이를 위해 실험실에서 배양된 세포나 실험 동물에 자극을 주고 그에 대한 신경 신호의 변화를 측정하는 실험들이 진행되어 왔다. 배양된 세포의 경우, 인간 뇌의 복잡한 구조와 다양한 기능을 실험 동물만큼 모사하기 어려운 단점이 있기 때문에 이를 개선하고자 뇌 장기 유사체가 개발 되었다. 하지만, 기존의 신경 신호 측정용 디바이스는 침습적이거나 측정 영역이 제한적이기 때문에 뇌 장기 유사체의 신경 신호를 측정하기가 적합하지 않다는 문제가 있다. 본 학위 논문에서는 비침습적으로 뇌 장기 유사체의 신경 신호를 넓은 영역에 걸쳐 측정하기 위한 신경 전극을 개발 및 제안하였다. 투명하고 유연한 폴리이미드에 구불구불한 패턴을 적용하여 신축성을 높였으며 두 영역으로 나누어서 각각 뇌 장기 유사체의 아랫면과 옆면의 신호를 측정할 수 있도록 하였다. 실험용 쥐의 뇌를 적출한 뒤에 제작한 신경 전극 위에 올려서 신경 신호를 측정하는데 성공하였지만, 전극과 세포의 접촉이 좋지 않아서 배지가 있는 상태에서 측정하지 못한 한계가 있다.
