The advantages of microfluidics include reducing expense with smaller reagents, inducing faster reactions, and precisely controlling samples with small sizes. Therefore, it has been extensively studied in biological, chemical and medical systems. Characteristics of fluids can be divided into different regions of Reynolds numbers. In Stokes flow (Re<1), flowing particles simply follow the streamlines. In the intermediate Re (~1 < Re < ~100) between Stokes and turbulent regimes, particles are affected by inertial lift forces and migrate across the streamlines. Then, particles are focused into equilibrium positions (or focusing positions). This phenomenon is called inertial focusing. Using inertial focusing, the location of particles can be manipulated within the microchannel without external forces.The number and configuration of focusing positions are affected by the channel structures. In this dissertation, we fabricated various microchannels using MEMS and investigated the relation between fluidic phenomena and channel structures. First, we elucidated the basics of inertial microfluidics in Chapter 1. In chapter 2, we investigated inertial focusing in non-rectangular cross-section channels (half-circle, triangle) and suggested the method for particle manipulation using various cross-section shapes. In chapter 3, we explained the change of focusing positions in triangular channel depending on particle size and Re, and presented the channels for size-based separation. In chapter 4, we observed the tendency of inertial focusing in accordance with particle size, Re and angles. In chapter 5, we presented the 3D curved triangular channels consisted with thin PDMS film with high flexibility. Then, we investigated the alteration of stable focusing positions by Dean flow formation and validated that curved triangular channel can be applied to both sheath-less flow cytometry and rare cell separation. In chapter 6, we suggested the novelty of research and future prospective of inertial microfluidics.
미세유체는 적은 양의 시약을 사용해 비용 절감이 가능하고, 빠른 반응속도를 유도하며, 작은 크기의 샘플을 정확하게 조작할 수 있다는 장점이 있어 생물학, 화학, 의학 분야에서 많이 연구되고 있다.유체의 특성은 레이놀즈수(Reynolds number)에 따라 구분되는데, 스톡스 흐름(Stokes flow, Re < 1) 영역의 유체에서 입자는 단순히 유선을 따라 흐른다. 스톡스 흐름과 난류(Turbulent flow) 사이의 층류 흐름(Laminar flow)에 해당하는 특정 영역의 레이놀즈수(~1 < Re < ~100)에서는 입자가 관성력(Inertial lift force)에 영향을 받아 유선에 수직인 방향으로 움직이고 힘의 평형점(혹은 집중점, Focusing position)에 집중된다. 이러한 현상을 관성집중(Inertial focusing)이라고 한다. 관성집중 현상을 이용하면 외부 힘없이 간단한 방법으로 채널 내의 세포와 미세입자의 위치를 원하는 곳으로 조절할 수 있다.입자 집중점의 위치와 개수는 채널 구조를 이용하여 조작할 수 있다. 본 학위논문에서는 MEMS 기술을 사용하여 다양한 구조의 미세채널을 제작하고, 유체역학적 현상 변화와 미세채널의 구조 관계를 이해하는 연구를 진행하였다. 먼저 Chapter 1에서는 관성미세유체와 관련된 현상에 대해 설명하고, Chapter 2에서는 반원, 삼각형 단면의 미세유체 채널을 사용하여 채널 단면 모양에 따른 입자의 거동 변화와 채널 단면 모양을 이용한 입자조작기술을 소개하였다. Chapter 3에서는 삼각형 채널에서 입자 크기와 레이놀즈수에 따른 입자 거동 변화에 대해 서술하고, 이 현상을 이용해 개발된 크기 기반 입자 분리 채널을 소개하였다. Chapter 4에서는 다양한 각도의 삼각형 채널에서 입자 크기, 레이놀즈수에 따라 유체역학적 변화를 관찰하고 현상 해석에 관한 내용을 기술하였다. 마지막으로 Chapter 5에서는 얇은 PDMS film를 사용하여 제작한 유연성이 높은 3D 곡면 삼각형 채널을 소개하였다. 그리고 삼각형 곡면채널에서 형성되는 딘 흐름(Dean flow)에 의한 안정된 입자 집중점의 위치를 변화를 관찰하고 곡면삼각형 채널이 조건에 따라 다양한 분야에 적용 가능함을 보였다. Chapter 6에서는 본 연구의 중요성과 관성미세유체 연구의 나아갈 방향에 대해 기술하였다.
