Part 1. Identification of 4-Phenoxyquinoline Based Inhibitors for L1196M Mutant of Anaplastic Lymphoma Kinase by Structure-Based Design
Dysregulation of anaplastic lymphoma kinase (ALK) has been detected in non-small cell lung cancer (NSCLC) in the form of EML4-ALK fusion. Secondary mutations opposing activity of the first generation ALK inhibitor crizotinib came into existence. In this study, I report 4-phenoxyquinoline-based inhibitors that overcome crizotinib resistance to ALK L1196M. The protonation of 4-aminoquinoline core could interrupt the ability the N atom of quinoline to act as a hydrogen bond acceptor; therefore, the pKa and calculated ionization pH values of relevant pyridine-based core moieties were carefully analyzed. Replacement of amine linkage with ether resulted in single-digit nanomolar range inhibitors.
Part 2. Discovery of Fluorescent 3-Heteroarylcoumarin Derivatives as Novel Inhibitors of Anaplastic Lymphoma Kinase
Altered expression or hyperactivation of anaplastic lymphoma kinase (ALK) is one of the main oncogenic drivers in non-small cell lung cancer. Using structure-based design and in vitro enzyme assays, I identified 3-heteroarylcoumarin as a new template for the development of novel fluorescent ALK inhibitors. Molecular simulation provided structural insights for the design of 3-heteroarylcoumarin derivatives, which were easily prepared through efficient synthetic approaches including direct C-H cross coupling. Importantly, these coumarin-based ALK inhibitors are suitable for a cell imaging tool (ALK/$IC_{50}$ = 0.51 $\mu M$, $\lambda_{emi}$ = 500 nm, $\phi_F$ = 0.29).
Part 3. Studies on Effective Dual Inhibition of mAChR M3 and CK1$\alpha$ to Suppress Active Signaling Pathway of ERK1/2 in Cancer Proliferation
Low subtype selectivity of mAChR agents induces serious problems in cancer treatment; therefore, securing selectivity became a debated clinical challenge. In this study, I proposed dual inhibition of specific mAChR subtype and the subtype receptor kinase to obtain the selectivity of inhibition of cell proliferation signaling.
Part 1. 구조 기반 설계를 통한 4-페녹시퀴놀린 구조체의 역형성 림프종 인산화효소 L1196M 돌연변이 억제제 개발
역형성 림프종 인산화효소는 융합된 형태의 과발현 현상으로 비소세포성 폐암을 포함한 여러 암을 유발하는 인자로 알려져 있으며, 1세대 억제제 처리시 발생하는 2차 돌연변이가 새로운 문제로 대두됨에 따라 돌연변이 형태를 억제할 수 있는 표적 항암제를 개발하기 위하여 연구를 시작하였다. 이번 연구에서는 역형성 림프종 인산화효소의 게이트키퍼 돌연변이를 억제하는 4-페녹시퀴놀린 구조체를 개발하여 1세대 약물의 내성을 극복하였다. 또한, 이온화 수소이온지수 계산값을 분석하여 체내 산성지수에서 4-아미노퀴놀린의 프로톤 부가 현상을 관찰하고, 리간드의 결합을 강화하기 위해 에테르 연결고리를 사용하여 억제 효과가 증대됨을 확인하였다.
Part 2. 새로운 역형성 림프종 인산화효소 억제제로서의 형광성 3-헤테로아릴쿠마린 구조체 개발
구조 기반 약물 디자인을 통하여 3-헤테로아릴쿠마린 구조체를 역형성 림프종 인산화효소의 형광성 억제제로 개발하였다. 분자 시뮬레이션 실험을 통해 적합한 변화를 유도하여 억제 활성을 증대시켰으며 각 물질에 대한 광 물리적 특성 또한 유지되는 것을 확인하였다. 또한 공초점 형광 현미경 관찰을 통해 세포 내 형광을 관찰할 수 있었으며 이를 통하여 3-헤테로아릴쿠마린 물질을 이용한 세포 이미징의 가능성을 확인하였다.
Part 3. 무스카린성 아세틸콜린 수용체 M3 와 카제인 키나아제 1$\alpha$의 이중 억제를 통한 암 확산 과정의 세포외 신호조절 인산화효소 신호 억제에 대한 연구
G 단백질 연결 수용체에 속하는 무스카린성 아세틸콜린 수용체는 하위 유형에 따라 각기 다른 기전을 통해 다양한 조절 기능을 하는 것으로 알려져 있다. 그러나 부적절한 활성이 관찰 되었을 때, 무스카린성 아세틸콜린 수용체 조절을 위한 물질이 하위 유형 선택성이 낮을 경우 심각한 부작용을 나타내는 것으로 알려져 있다. 이와 같은 문제를 극복하기 위하여 이 연구에서는 해당 하위 유형의 무스카린성 아세틸콜린 수용체와 이의 키나아제를 이중 저해함으로서 신호 억제 선택성을 증대시키고자 하였다.
