As fossil fuel depletion and climate change has been serious, lignocellulosic biomass has attracted attention. Lignocellulosic hydrolysates contain a large amount of glucose and xylose, but due to the rare strains that can efficiently use xylose, it was rarely used as actual sources. In this study, this problem was solved by using the strain that has already been adaptively evolved in 3.6 g/L xylose. In addition, a gamma aminobutyric acid, GABA, production pathway was constructed in the ALE strain. GabT, GabD, and GabP genes were deleted to enhance GABA production. However, when high concentration glucose was present, xylose was rarely consumed. Thus, the new adaptive evolution was performed under the relatively high concentration of xylose. The newly adaptively evolved strain showed a better growth curve than the parent strain under glucose and xylose mixed conditions. If the newly discovered adaptive evolution strain is further engineered, a cell factory that produces gamma aminobutyric acid while simultaneously consuming glucose and xylose in lignocellulosic hydrolysates will be constructed.
화석연료 고갈 가능성과 기후 변화가 심화됨에 따라 목질계 바이오매스가 주목을 받아왔다. 목질계 바이오매스는 포도당과 자일로오스를 다량 함유한 자원이지만, 효율적으로 자일로오스를 이용할 수 있는 균주가 드물어 실제 원료로 사용되기 어렵다. 본 연구에서는 3.6 g/L 자일로오스에 적응 진화된 균주를 사용해 이러한 문제를 해결하고자 한다. 또한 해당 균주에 감마 아미노부틸산 생산 경로를 구축하고, GabT , GabD , GabP 유전자를 제거해 감마 아미노부틸산 생산을 증대시켰다. 하지만 고농도의 포도당이 존재할 경우, 자일로오스를 전혀 소모하지 못하는 문제가 확인되어, 상대적 고농도 자일로오스 조건에서 적응진화를 수행했다. 새롭게 적응 진화된 균주는 포도당 및 자일로오스 혼합 조건에서 모균주보다 우수한 성장 곡선을 보였다. 새롭게 발굴된 적응진화 균주를 추가적으로 개량한다면 목질계 당화액 내의 포도당과 자일로오스를 동시 소모해 감마 아미노부틸산을 생산하는 세포공장을 만들 수 있을 것으로 기대된다.
