The two-dimensional (2D) culture method has been used for cancer research because of its ease of use, but has disadvantages that cannot mimic characteristics of actual tumors. To overcome this, many efforts have been made to generate three-dimensional (3D) cancer cell spheroids that can mimic cancer cell characteristics. However, the culture methods already developed have challenges in terms of controlling the characteristics of cancer cells and cancer stem cells. Since the trait of cancer cells is known to be greatly influenced by the surrounding environment in which the cells are cultured, it is possible to control the characteristics of cancer cells through surface modification of the culture substrate. Therefore, this study intends to confirm the change in cell morphology and properties according to the surface energy by modifying the surface with the low surface energy-polymer and the high surface energy-polymer and their copolymers. We fabricated a series of surface energy-regulated polymer culture dishes via initiated chemical vapor deposition (iCVD). We confirmed that according to surface energy, the pattern of the adsorbed protein was changed, and consequently the change in cell morphology. In particular, it was uncovered that the adsorbed albumin plays a pivotal role in changing cell characteristics. Also, we identified that the degree of conformational change in the secondary structure of albumin increased as the surface energy decreased. This research could provide a new approach to the development of cancer cell spheroids platform that can mimic the tumor environment.
2 차원 배양법은 용이성 때문에 암 세포 연구에 사용되지만 실제 암세포 특성을 모사하기 힘들다는 단점이 있다. 이를 극복하기 위해, 암세포 특성을 잘 모방할 수 있는 3차원 암세포 스페로이드를 개발하기 위한 많은 노력들이 이루어지고 있다. 그러나, 이미 개발된 배양법들은 암세포의 특성제어 및 암줄기세포의 특성이 미미하다는 단점이 있다. 암세포의 형질은 세포가 배양되는 주변환경에 의해 크게 영향을 받는다고 알려져 있기 때문에 배양 기판의 표면개질을 통해 암세포의 특성제어가 가능하다. 따라서 본 연구에서는 낮은 표면 에너지를 지닌 고분자와 높은 표면 에너지를 지닌 고분자, 그리고 이들의 혼성고분자를 통해 표면을 개질함으로써 표면 에너지에 따른 세포 형태 변화와 이에 따른 세포의 특성 변화를 확인하고자 한다. 개시제를 이용한 화학 기상 증착 (iCVD)을 통해 표면 에너지를 조절한 일련의 고분자 배양 접시들을 제조하였다. 표면에너지에 따라 흡착단백질의 양상이 달라지고, 이에 따른 세포 형태 변화가 확인되었다. 특히 흡착된 알부민이 세포 특성변화에 중추적인 역할을 함을 발견하였고, 표면 에너지가 감소함에 따라 알부민 이차 구조의 변화 정도가 증가함을 확인하였다. 해당 연구는 종양 환경을 모방할 수 있는 암세포 스페로이드 플랫폼 개발에 대하여 새로운 접근 방식을 제공해줄 수 있을 것이다.
