$Ventral anterior homeobox1$ ($VAX1$) is a homeodomain transcription factor, which is known for its role in the development of vertebrate ventral forebrain. $VAX1$ mutations in human and mice cause various malformation of ventral midline brain structures including anterior commissure, corpus callosum and optic chiasm. Previously, it was reported that Vax1 not only regulates transcription of genes in the brain areas but also promotes retinal axon growth as a translational regulator via the intercellular transfer. However, it still remains unknown about the physiological importance of the intercellular Vax1 transfer in the development. To investigate this, we generated Vax1AA mutant mice, in which conserved lysine and arginine (KR) residues that bind to heparin sulfate proteoglycan (HSPG) of retinal axons are replaced with two alanines (AA). We then examined the brain and eye development of mice at six different Vax1 gene contents. The $Vax1^{-/-}$ and $Vax1^{-/AA}$ mice failed to form the midline structures of forebrain and died perinatally due to the palate cleft. The $Vax1^{AA/AA}$ mice are also lacked of the commissures but survive by having normal palate. The optic chiasm was not formed in $Vax1^{AA/AA}$ mice, and thus the optic nerve extended to the ipsilateral brain areas. Visual information was processed properly in the retina of $Vax1^{AA/AA}$ mice, however it could not trigger vision-triggered behavior properly. Together, we propose that the intercellular transfer of Vax1 is necessary for the formation of optic chiasm and the achasmatic $Vax1^{AA/AA}$ mice can be used to understand the spatial resolution of mouse vision
호메오도메인 전사인자 중 하나인 VAX1 은 척추동물의 전뇌 발달과정에서 중요한 역할을 한다고 알려졌다. VAX1 유전자의 변이는 사람과 생쥐에게서 공통적으로 비정상적인 전뇌 중심선 구조를 나타낸다. 앞선 연구에서 Vax1 은 뇌에서 전사인자로서의 역할 뿐 아니라, 세포간 이동현상을 통해 번역 조절자로서 망막세포의 축색돌기 성장을 조절한다는 것이 밝혀졌다. 하지만, Vax1의 세포 간 이동현상이 나타내는 생리학적 중요성에 대해서는 아직 밝혀지지 않았다. 이러한 문제점을 해결하기 위해 우리는 Vax1 이 세포간 이동하는데 있어서 헤파린과 결합하는 부위인 라이신과 아르기닌(KR)을 모두 알라닌(AA) 으로 치환한 Vax1AA 변이 생쥐를 만들었다. Vax1AA/AA 생쥐는 Vax1-/- 생쥐 와는 달리 정상적인 입천장 구조를 가졌으나 시신경 교차는 형성되지 않았다. 또한, Vax1AA/AA 생쥐는 망막에서의 기능은 정상적인 모습을 보이지만 시신경교차가 형성되지못하여 시신경이 동측 뇌 영역으로만 뻗어나갔다. 그로인해 시각정보전달에 역전이 일어나 시각 반응에 따른 적절한행동을 하지못하였다. 따라서 우리는 본 연구를 통해 Vax1의 세포간 이동현상이 시신경교차 형성에 있어서 필수적이며, 더 나아가 이 생쥐를 통해 시각처리과정에 대한 이해를 높이는데 이용할 수 있다.
