Mucosal-associated invariant T (MAIT) cells, the most abundant innate-like T cells in human liver, can be activated by MHC class I-related molecule 1 (MR1)-dependent TCR stimulation or by cytokines in the absence of TCR stimulation. However, whether MAIT cells can exert TCR/MR1-independent cytolytic activity remains unclear. Here, I examined the mechanisms underlying TCR/MR1-independent innate-like cytotoxicity of cytokine-activated liver MAIT cells. I also investigated the phenotype and function of MAIT cells from patients with viral hepatitis. IL-15-stimulated MAIT cells exerted granzyme B-dependent innate-like cytotoxicity in the absence of TCR/MR1 interaction. PI3K–mTOR signaling, NKG2D ligation, and CD2-mediated conjugate formation were all critically required for this IL-15-induced innate-like cytotoxicity. MAIT cells from patients with acute hepatitis A (AHA) exhibited activated and cytotoxic phenotypes with higher NKG2D expression. The innate-like cytotoxicity of MAIT cells was significantly increased in patients with AHA and correlated with serum alanine aminotransferase levels. In summary, the results demonstrate that liver MAIT cells activated by IL-15 exert NKG2D-dependent innate-like cytotoxicity in the absence of TCR/MR1 engagement. Furthermore, the innate-like cytotoxicity of MAIT cells is associated with liver injury in patients with AHA, suggesting that MAIT cells contribute to immune-mediated liver injury in liver disease.
점막연관 불변사슬 T 세포는 사람 간에 풍부하게 존재하는 선천면역유사 T 세포의 일종으로, T 세포 수용체/항원 제시 분자 MR1을 통한 자극 또는 T 세포 수용체-비의존적 사이토카인 자극으로 활성화될 수 있다. 하지만, 현재까지 점막연관 불변사슬 T 세포가 T 세포 수용체-비의존적 선천면역유사 세포독성을 발휘할 수 있는지에 대해선 알려진 바가 없었다. 이에 본 연구에서는, 간 이식 기증자의 간 관류액에서 T 세포들을 분리하여 사람 간 점막연관 불변사슬 T 세포에 대한 다양한 사이토카인의 효과를 유세포분석을 통해 확인하였고, 바이러스성 간염 환자의 혈액과 간 조직을 얻어 점막연관 불변사슬 T 세포의 표현형을 분석하였다. 그 결과, 인터루킨-15에 의해 활성화된 점막연관 불변사슬 T 세포가 granzyme B에 의해 매개되는 T 세포 수용체-비의존적 선천면역유사 세포독성을 발휘하는 것을 확인하였다. 이러한 인터루킨-15에 의해 유도되는 세포독성에서 PI3K–mTOR 신호전달경로, NKG2D의 결착, CD2에 의해 매개되는 세포 부착 모두 필수적인 역할을 함을 확인하였다. 이에 더해, 점막연관 불변사슬 T 세포의 NKG2D 발현 정도와 선천면역유사 세포독성이 급성 A형 간염 환자에서 모두 유의미하게 증가하였고, 이들의 증가는 환자의 혈청 alanine aminotransferase 수치와 양의 상관관계를 보였다. 이는 NKG2D에 의한 점막연관 불변사슬 T 세포의 선천면역유사 세포독성이 급성 A형 간염에서 발생하는 간 손상에서 중요한 역할을 할 수 있음을 시사한다. 요약하면, 인터루킨-15에 의해 활성화된 점막연관 불변사슬 T 세포는 NKG2D-의존적 선천면역유사 세포독성 기능을 가지고, 이러한 세포독성이 간 질환에서의 면역매개 간 손상에서 중요한 역할을 할 것으로 사료된다.
