Modulating proteins activities via light-activated photoreceptors offered unexpected advance in the field ofscience. Especially universal downstream signaling molecule, $Ca^{2+}$, may provide insights on the dynamic natureof cellular behaviors and result in the comprehension of higher order outcomes due to extensive usages tointerconnect disparate cellular networks. Previously we designed $Ca^{2+}$-selective optogenetic tool by activatingendogenous CRAC channel has been applied to manipulate $Ca^{2+}$ activities in many cellular systems and even invivo system. Blue-light driven oligomerization of cryptochrome2 (CRY2) enable to form active STIM1conformation and subsequently promote activation of endogenous CRAC channels, in turn, elevating $[$Ca^{2+}$]_i$.However, the low penetration property of blue light-responsive photoreceptors is considered inadequate to use invivo without inserting optic fiber insertion. Here, we present an ultra-light sensitive optogenetic $Ca^{2+}$ modulator,named “monSTIM1” developed by engineering cryptochrome2 to control $Ca^{2+}$ signaling in the brain of awakemice through non-invasive light delivery. Through expressing monSTIM1 in mice brain, we are able to induce $Ca^{2+}$-dependent immediate early gene, c-Fos expression without any alteration of intact mice brain. Furthermore, $Ca^{2+}$ relative behavior of mice can be affected through non-invasive activation of monSTIM1 demonstrating itsability to control $Ca^{2+}$ signaling in the brain of awake mice through non-invasive light delivery.
빛에 의해 활성화 되는 광 수용체 단백질을 이용하여, 원하는 단백질 기능을 조절하는 것은 과학 분야에서예기치 않은 진보를 가져왔다. 특히 세포 내의 신호 전달을 일으키는 칼슘 이온은 세포 내 상호적 연결의전반적인 이해를 가져올 수 있어 세포 내 칼슘 이온을 선택적으로 투과 시키는 CRAC 채널을 빛으로활성화하는 광 활성 기술을 개발하여 빛을 이용해 많은 세포 시스템 및 생체 내 시스템에서 칼슘 이온을유입시키고 활동을 조작할 수 있었다. 이 기술은 아기장대에서 추출한 크립토크롬2 단백질이 청색 빛에 의한올리고머를 형성하게 되고, 다형체가 형성되었을 때 STIM1 기능이 활성화를 통해 세포 내 보편적으로 발현되어 있는 칼슘 채널인 CRAC 채널의 활성화를 촉진시켜 세포 내 칼슘 이온 농도를 상승시킨다. 그러나,이러한 기술은 생체 내 투과성이 떨어져 광섬유 삽입을 하지 생체 모델에 이용하는 것이 부적절하다. 이를극복하기 위해 우리는 크립토크롬2 단백질의 아미노산을 치환하여 청색광에 더 민감하게 반응하는 monSTIM1을 개발하였으며, 비 침습적인 빛 전달을 통해 깨어있는 쥐의 뇌에서 칼슘 신호 전달을 조절이 가능한 것을확인하였다. 이를 통해 우리는 비 침습적인 monSTIM1의 활성을 유도하여 칼슘과 관련된 생쥐의 행동 변화를유도한다는 것을 통해 칼슘이 뇌의 여러 세포 종류, 다양한 부위에서의 기능을 연구할 수 있을 것이다.
