In the liver, 80% of hepatocellular carcinoma (HCC) development in a microenvironment is characterized by chronic injury, inflammation, as well as fibrosis. However, certain cellular mediators which are responsible for the high risk to promote HCC are not well studied, unfortunately. Here we investigated the hypothesis that hepatic stellate cells (HSCs) promote HCC, a consequence of recruiting of myeloid-derived suppressor cells (MDSCs) which known as immunosuppressive cells. In the study of HCC mouse models and analyses of HCC patient specimens, activated HSCs near hepatic tumor site dramatically induced infiltration of MDSCs. After MDSCs interacting with HSCs accelerated tumor progression by inhibiting proliferation of cytotoxic T lymphocyte via arginase-I dependent manner. Our data suggest that the activation of MDSCs by HSCs might contribute to immune suppression during hepatic tumor progression. Therefore, the interaction of activated HSCs and MDSCs might be the new therapeutic targets for prevention of HCC
간암의 발병원인은 80% 이상이 만성적인 염증과 같은 간 손상으로부터 시작이 되며, 이러한 현상은 만성 염증, 간 섬유화와 같은 초기 질환으로부터 시작이 된다. 하지만, 간암 발생에 대한 종양 미세환경에서 어떤 특정 면역 세포들간의 상호작용이 간암 발생을 촉진하는 지에 대해서 잘 알려지지 않고 있다. 간암 미세 환경에서 활성화 된 간성상세포가 면역 억제 조절 세포로 잘 알려진 미성숙 골수 억제 세포를 유도하여 간암 발생을 더욱 촉진할 것이라는 가설 하에, 간암 유도 모델뿐만 아니라, 간암 환자 유래의 샘플 연구를 통해 확인한 결과, 종양 주위에 활성화 된 간성상 세포가 미성숙 골수 억제 세포를 간암 미세 환경으로 이동 및 유도한다는 것을 확인 하였다. 그리고, 이 두 세포간의 상호 작용을 통해 종양 억제 세포인 T 세포의 증식을 억제하며, 이러한 현상은 간성상 세포의 retinoid 분비에 따른 미성숙 골수 세포의 arginase-1 매개로 이루어 진다는 것 또한 확인할 수 있었다. 그렇기에 활성화 된 간성상 세포와 미성숙 세포간의 상호작용이 간암 발생을 촉진하기에, 이 두 세포간의 상호 작용을 억제한다면 간암 발생에 대한 새로운 치료법의 가능성을 확인해 주는 것이다.
