Biofuel market has been progressively grown including the production of biodiesel from feedstock such as soybean oil to reduce the proportion of fossil fuels. Consequently, removing the excess glycerol produced as a by-product formed during the biodiesel transesterification surfaced as a new problem. Using glycerol as a carbon source for microorganisms to produce value-added products is suggested as a solution. For example, there were efforts to utilize Escherichia coli for the conversion of glycerol to high value product such as $\gamma$-aminobutyric acid. Here, E. coli W underwent 1,300 generations of adaptive laboratory evolution in glycerol minimal medium to improve its GABA production from glycerol. The parental strain obtained several mutations including glycerol kinase GlpK during the adaptive evolution. Through literature and phenotype study, we characterized the mutant GlpK as the key mutation. In addition to the biochemical characterization of the mutant enzyme, transcriptome analysis spotted change in crp regulons including GABA shunt involved genes gabT and gabD. We anticipated that such phenomenon would be advantageous for GABA production, and achieved 0.310 g L-1 of GABA from 2 g L-1 glycerol, the highest GABA titer from glycerol using E. coli solely.
대두 기름과 같은 바이오원료로부터 바이오디젤을 생산을 포함하여 바이오연료 시장은 화석연료를 대체하기 위해 꾸준히 성장해왔다. 바이오디젤을 생산하는 과정에서 바이오디젤 전이에스테르화 반으으이 부산물인 글리세롤이 대량 생산되었고 이를 처리하는 것이 새로운 문제로 대두되었다. 이 글리세롤을 미생물의 탄소원으로 사용하여 감마-아미노부티르산과 같은 고부가가치 물질로 변환하는 것이 하나의 해결책으로 제안되었다. 본 연구에서는 E. coli W를 글리세롤 조건에서 약 1,300 세대의 실험실 적응진화를 진행하였다. 차세대 유전체 시퀀싱과 돌연변이 실험을 통해 글리세롤 키나제 GlpK를 적응진화 도중에 발생한 주요 돌연변이로 지정하였고 해당 효소의 생화학적 분석과 전사체 분석을 통해 적응진화를 마친 균주 CY02가 감마-아미노부티르산을 분해하는 효소인 GabT와 GabD의 유전자 발현이 억제된 것을 확인하였다. 해당 효소의 발현이 억제된 이 균주는 감마-아미노부티르산을 고생산 할 수 있을 것이라 예측하였고 실제로 대조군에 비해 글리세롤부터 향상된 감마-아미노부티르산 생산성을 확인할 수 있었다.
