We herein developed a novel isothermal method to detect platelet-derived growth factor (PDGF) utilizing target-induced nicking and extension chain reaction system-based amplification (NESBA). In principle, the binding of PDGF to the aptamer strand (AS) that was priorly hybridized with template strand (TS) liberates TS which then initiates the following NESBA system, leading to the exponential amplification of RNA amplicons that are monitored by molecular beacon (MB). With this method, we sensitively determined a model target, PDGF-BB, down to 6.48 nM with high selectivity against non-specific analytes including different types of PDGFs. More importantly, we also demonstrated the capability of this sensor to reliably analyze PDGF-BB in the human serum sample, verifying its practical applicability to the development of point-of-care (POC) device for cancer diagnosis.
본 학위논문에서는 절단 및 중합 연쇄 반응 시스템 기반 증폭 기술을 활용한 혈소판 유래 성장인자의 신개념 등온 검출 방법을 개발하였다. 본 기술은 혈소판 유래 성장인자가 있을 경우, 주형가닥과 상보적으로 결합된 압타머가닥에 결합하여 주형가닥을 방출시킨다. 방출된 주형가닥은 절단 및 중합 연쇄 반응 시스템을 구동시켜 기하급수적으로 형광물질이 수식된 분자 비콘과 반응하는 RNA 산물을 생성한다. 본 기술을 통해, 혈소판 유래 성장인자를 6.48 nM의 낮은 검출 한계로 검출하였으며, 혈소판 유래 성장인자의 다른 종류를 포함하는 비특이적인 피분석물로부터 높은 선택도를 확보하였다. 최종적으로 사람의 혈청에서 본 기술이 혈소판 유래 성장인자를 성공적으로 검출하여 암의 현장진단 기술에 적용 가능한 실용성을 증명하였다.
