Sequential firing patterns of place cells along a spatial trajectory are often replayed during sharp wave ripple (SWR) epochs during awake immobility. These brief sequential burst of spikes can occur both in forward and reverse directions. The functional roles and mechanisms underlying these awake replays are not clear. To obtain better insight into this matter, we examined hippocampal replays in a novel spatial sequence memory task. Reward enhances the rate of reverse replays, but it increases the fidelity of forward replays for recently-travelled as well as other alternative trajectories heading toward a rewarding location. This suggests roles of forward and reverse replays in reinforcing representations for all potential rewarding trajectories. We also found more faithful reactivation of place cell sequences for upcoming rewarding trajectories than already rewarded trajectories in forward replays. This suggests a role of forward replays in preferentially reinforcing representations for high-value trajectories. Our results suggest hippocampal forward and reverse replays may contribute to constructing a map of potential navigation trajectories and their associated values ('value map') via distinct mechanisms.
공간에 대한 해마 위치세포들의 발화는 수면이나 휴식 중 순차적으로 재생된다는 것이 알려져 있다. 또한 이들 해마 위치 세포의 순차적 발화양상은 순방향 및 역방향으로 재생된다. 그러나 이러한 순방향 및 역방향 재생의 원리와 기능은 아직 밝혀지지 않았다. 본 연구에서는 네 개의 경로 중 하나를 기억하여 선택하여야 보상이 주어지는 공간 기억 과제를 새롭게 고안하여, 동물이 과제를 수행하는 동안 해마 위치세포의 순차적 발화 양상과 재생 패턴을 기록하고 분석하였다. 그 결과 보상이 주어진 시행에서는 모든 경로에 대해서 역방향 재생에 있어서는 위치세포 발화의 재생 빈도가 증가하였고, 반면에 순방향 재생에 있어서는 위치세포 발화의 재생 정확도가 증가하였다. 또한 순방향 재생에 있어서 보상이 주어졌던 이전 경로에서 발화했던 위치세포보다 다음 번에 보상이 주어질 경로에서 발화하는 위치세포들의 재활성화가 강도 높게 관찰되었다. 이러한 결과들은 해마 위치세포 발화의 재생은 잠재적 탐색 경로와 그와 연관된 효용가치(효용가치 지도)의 표상에 기여하며, 이 과정에 순방향 및 역방향의 재생이 각기 다른 방법으로 기여함을 시사한다.
