NF- $\kappa$ B signaling through both canonical and non-canonical pathways plays a central role in cell proliferation, inflammation and immune responses. NF- $\kappa$ B -inducing kinase (NIK) stabilization is a key step in activation of the non-canonical pathway and its dysregulation implicated in various hematologic malignancies. The tumor suppressor, p53, is an established cellular gatekeeper of proliferation. Abnormalities of the TP53 gene have been detected more than half of all human cancers. While the non-canonical NF- $\kappa$ B and p53 pathways have been explored for several decades, no studies to date have documented potential cross-talk between these two cancer-related mechanisms. Here, I demonstrate that p53 negatively regulates NIK in an miRNA-dependent manner. Overexpression of p53 decreased the levels of NIK, leading to inhibition of the non-canonical NF- $\kappa$ B pathway. Conversely, its knockdown led to increased levels of NIK, IKK $\alpha$ phosphorylation, and p100 processing. Additionally, miR-34b induced by nutlin-3 directly targeted the coding sequences (CDS) of NIK. Treatment with anti-miR-34b-5p augmented NIK levels and subsequent non-canonical NF- $\kappa$ B signaling. My collective findings support a novel cross-talk mechanism between non-canonical NF- $\kappa$ B and p53.
NF- $\kappa$ B 신호 전달은 전형적 경로와 비전형적 경로를 통해 세포 증식, 염증 및 면역반응에서 중요한 역할을 한다. NF- $\kappa$ B -inducing kinase (NIK)의 안정화는 비전형적 NF- $\kappa$ B 경로의 활성화를 위한 주된 단계이므로 NIK의 조절 장애는 다양한 혈액 암을 초래한다. 종양억제유전자인 p53은 세포 증식의 문지기유전자로 여겨지고 있고, 전체 암의 절반 이상에서 p53 유전자의 변이가 발견된다. 그동안 비전형적 NF- $\kappa$ B 경로와 p53 경로는 오랜 기간 연구되어왔으나 두 경로의 상호조절에 대한 연구는 미흡하다. 이 논문을 통해 p53이 miRNA 의존적 방법으로 NIK을 음성 조절함을 발견하였다. p53의 과발현은 NIK의 발현을 감소시켰고 결과적으로 비전형적 NF- $\kappa$ B 경로를 억제하였다. 또한 nutlin-3 처리로 증가된 miR-34b는 NIK 전사체의 단백질 암호화 서열을 직접적으로 표적으로 한다. 더 나아가 miR-34b-5p 억제제의 처리는 NIK의 발현 및 비전형적 NF- $\kappa$ B 신호전달을 증대시켰다. 결국, 이 논문은 두 비전형적 NF- $\kappa$ B와 p53 경로의 새로운 상호조절 매커니즘을 제시한다.
