The retina is a light-sensitive tissue that converts the light into visual stimulus. Once the retina is affected by pathological change, it is hardly reversible and can lead to vision loss and even permanent blindness. Understanding the pathogenesis of retinal diseases is greatly facilitated by utilizing small animal models, such as rodent model. For investigating pathophysiology and drug development in rodent model, it is essential to visualize structure and vasculature of retina tissue.
Optical coherence tomography angiography (OCTA) is a non-contact and non-invasive imaging modality visualizing 3-dimensional microstructure and microvasculature, in-vivo. In this study, high-speed OCTA for imaging rodent retina was developed and longitudinal analysis of vascular changes in rodent disease models were analyzed. Also, OCTA imaging of the rodent retina for volumetric visualization of microvascular blood flow were demonstrated and hemodynamic changes were investigated.
시각 형성의 중추 기관인 망막에 병리적 변화가 발생할 경우 시력이 감소하고 경우에 따라 영구적으로 시력을 상실한다. 이러한 망막질환의 병리생리학적 기전 및 약물 및 시술에 대한 반응을 연구하는 것은 중요하지만 사람을 대상으로 연구를 진행하는 것은 많은 어려움이 존재하기 때문에 실험 동물, 특히 설치류 동물을 활용한 전임상 연구를 선행한다. 이 때, 생체 내 망막을 영상 촬영하는 것은 질병의 발병 과정을 관찰하거나 치료 약물의 효과 및 경과를 보기 위해서 필수 불가결하다. 본 연구에서는 설치류 망막의 3차원 미세구조 및 미세혈관 구조를 비접촉, 비침습적으로 이미징할 수 있는 OCTA(Optical coherence tomography angiography) 시스템을 개발하여, 다양한 설치류 질병 모델에서 미세구조 및 미세혈관 구조의 형태학적 변화를 관찰하고 분석한다. 또한 OCTA를 활용하여 설치류 망막의 미세혈류속도를 측정하여 중증질환에서 혈류역학적 변화에 대해 관찰하고 분석한다.
