Surface-enhanced Raman scattering (SERS) is non-destructive and ultra-sensitive analytical tool using localized surface plasmon resonance on the surface of metal nanostructures. However, metal surfaces are prone to contamination by irreversible adsorption of large adhesive molecules, such as proteins, in the biological fluid, which hinders the access of small target molecules on the surface of the metal. The pretreatments such as centrifugation, dialysis are necessary for exclusion of large adhesive molecules. During pretreatment, the loss of target molecules and consumption of money and time is inevitable. In this thesis, I focus on direct detection of small molecules in biological fluids without pretreatment. Using microfluidics, I design the metal nanoparticle loaded the semi-permeable hydrogel. Metal nanoparticle makes SERS-active site and hydrogel render size or charge selectivity of molecules in the complex mixture.
표면 증강 라만 산란(SERS)은 금속 나노 구조 표면에서 비롯되는 국부적인 표면 플라스몬 공명을 이용한 비파괴적이고 초민감한 분자구조 분석방법이다. 생체유체 내에서 SERS 분석 시, 금속 표면은 유체내에 존재하는 단백질과 같은 분자의 비가역적 흡착으로 인해 금속 표면으로의 작은 타겟분자의 접근을 방해한다. 이에 금속 표면에 붙기 쉬운 단백질을 배제하기 위해 원심분리, 투석과 같은 전처리 과정이 필요하고, 이 과정에서 타겟분자의 손실과 경제적인 소모가 불가피하다. 본 학위논문에서는 전처리 없이 생물학적 유체에서 크기가 작은 타겟분자를 직접 검출하기 위한 연구를 진행하였다. 미세유체소자를 사용하여 선택적인 투과성을 가지는 하이드로젤 내에 금속 나노 입자를 함유한다. 금속 나노 입자는 SERS 활성사이트를 만들고 하이드로젤은 복잡한 혼합물에서 분자의 크기 또는 전하에 따라 선택적인 투과를 하여 타겟분자만을 SERS 활성사이트에 흡착하게 하여 전처리 없이 타겟분자만의 검출을 가능하게 한다.
