Fas receptor (CD95) is known to lead both cell death and pro-survival outputs and be activated in numerous neurological diseases. Given its complexity, how the Fas signaling pathway operates dynamically has not yet been fully understood. Recent advances in optogenetics enable a thorough investigation of a signaling network which can also explain cellular responses or fates, including the phenomenon observed in various disorders. We here developed chemogenetically and optogenetically activatable Fas modules that operate in a physiologically plausible manner. Activation of Fas in immature neurons in the dentate gyrus induced mTOR activation and subsequent secretion of BDNF. Phosphorylation of Erk in neural stem cells was triggered upon prolonged Fas activation. When repetitively activated, this signaling network resulted in the proliferation of neural stem cells and a transient enhancement of spatial working memory in mice. Our work demonstrates a novel Fas signaling network in the dentate gyrus with optogenetic dissection as well as its consequences regarding adult hippocampal neurogenesis and memory increase.
Fas 수용체의 활성은 세포의 종류와 주어진 상황에 따라 세포 사멸과 증식을 모두 유도할 수 있는 복잡한 신호전달 네트워크를 유도한다. 다양한 뇌질환에서 Fas 수용체의 활성이 관찰되나, 해마에서 그 신호전달이 어떻게 조절되는지에 대한 연구는 이루어지지 않았다. 시공간적 조절이 가능한 방법을 통한 신호전달 경로의 연구는 다양한 생리학적 현상과 질병에서 관찰되는 현상에 대한 분자 수준의 해석을 가능하게 해준다. 이 연구에서 우리는 화학물질 및 청색광에 의해 활성화되는 Fas 수용체를 개발하고, 이것이 세포 내에서 생리학적으로 예측 가능한 결과를 유도할 수 있음을 보였다. 광유도 Fas 수용체를 생쥐 뇌의 해마 치아 이랑에 존재하는 미성숙 뉴런에 발현시키고 광조사 시간을 달리하는 실험을 통해, 우리는 미성숙 뉴런에서의 Fas 활성화가 해당 뉴런에서의 mTOR 신호전달 활성화와 뇌유래신경영양인자의 분비를 유도하며, 이를 통해 신경줄기세포에서 Erk 신호전달의 활성화가 순차적으로 일어난다는 것을 확인하였다. 또한, 이 신호전달 네트워크의 반복적인 활성화를 통해 신경줄기세포의 증식과 공간 운동 기억의 일시적 증가가 일어난다는 것을 밝혀내었다. 이를 통해 우리는 해마 치아 이랑에서 일어나는 새로운 Fas 신호전달 네트워크를 밝혔을 뿐 아니라 이것이 성체 신경 재생 및 기억 향상에 역할을 한다는 사실을 규명하였다.
