Numerous interactions in nature employ multivalent binding, where multiple ligands on one entity bind to multiple receptors on another. The avidities resulted in multivalent presentation of a ligand are orders of magnitude stronger than those for the corresponding monovalent interaction. Thanks to these unique collective properties, multivalent interaction is extensively used strategy to achieve high selectivity and affinity in researching area as well as in nature. But the detailed mechanism of these multivalent interactions and specific rule of binding is not revealed. Chapter 1 provides current understandings of multivalent interaction that kinetically approaches to the behavior and covers recent strategies for designing supramolecular protein assembly. Chapter 2 describes the changes of kinetic values ($k_{on} and k_{off}$) upon valency change of the protein-peptide pairs having different binding chemistry. By employing these multivalent interaction to nano-assembly, chapter 3 introduces a strategy to fabricate monodisperse protein assemblies; protein nano-prisms.
자연계의 많은 결합들은 다중 결합, 즉 하나의 개체에 존재하는 여러 개의 리셉터와 다른 개체에 존재하는 여러 개의 리간드가 동시에 결합하는 현상을 이용한다. 특이적인 일대일 결합이 반복됨에 따라, 다중 결합은 단일 결합들의 합을 뛰어넘는 강한 결합력을 이룩할 수 있다. 이러한 특성으로 인해, 다중 결합은 매우 특이적이고 강한 결합력을 요구하는 곳에서 다방면으로 이용되는 전략으로 자리 잡았다. 하지만 다중 결합의 메커니즘에 관한 연구는 제한적이다. 본 연구 논문에서는 다중 결합의 메커니즘을 운동역학적인 관점에서 살펴보고, 이를 이용해 새로운 단백질 조립체를 만드는 전략을 소개하고자 한다. 먼저, 서로 다른 방식으로 결합하는 단백질-펩타이드 결합 쌍이 결합 개수 별로 결합/해리 속도가 어떻게 달라지는지 관찰하였다. 그리고 다중 결합을 이용하여 단일 형태의 단백질 조립체를 만들 수 있는 전략으로서의 응용 가능성을 제시하였다. 본 연구는 단백질 간 다중 결합의 밸런시와 결합/해리 속도의 상관관계에 대한 이해를 넓혔으며, 다중 결합을 통해 단일 단백질 조립체를 만드는 전략을 제시하였다. 이를 바탕으로 추후 활발한 연구가 진행되기를 기대한다.
