Spatiotemporal control of gene expression or labeling is a valuable strategy for identifying functions of genes within complex neural circuits. Here, we developed a highly light-sensitive and efficient photoactivatable Flp recombinase (PA-Flp) that is suitable for genetic manipulation in vivo. The highly light-sensitive property of PA-Flp is ideal for activation in deep mouse brain regions by illumination with a noninvasive light-emitting diode. In addition, PA-Flp can be extended to the Cre-lox system through a viral vector-expressing Flp-dependent Cre-activation platform, thereby activating both Flp and Cre. Finally, we demonstrate that PA-Flp-dependent, Cre-mediated Cav3.1 silencing in the medial septum increases object-exploration behavior in mice. Thus, PA-Flp is a noninvasive, highly efficient, easy-to-use optogenetic module that offers a side-effect-free and expandable genetic-manipulation tool for neuroscience research.
시공간적으로 유전자를 발현하거나 표지를 하는 기술은 복잡한 뉴런의 회로 내에서 유전자의 기능을 연구하는데 있어 큰 가치가 있다. 우리는 빛에 반응하여 활성화되는 Flp 재조합 효소 (광활성 Flp) 를 개발하였고, 이를 이용해 살아있는 생체 내에서 유전자의 발현을 조작할 수 있도록 하였다. 빛에 매우 민감한 특성을 가진 광활성 Flp 기술은 발광 다이오드 (light-emitting diode)를 이용해 쥐의 깊은 뇌에까지 조사하는 방식으로 응용하여 사용 가능하다. 또한, 광활성 Flp 기술을 바이러스 벡터에 의해 조절되는 Flp 의존적인 Cre 활성 플랫폼으로 구축함을 통해 Cre-Lox 시스템까지 확장성 있는 사용을 제시하였다. 이를 활용하여, 광활성 Flp 의존적인, Cre에 의한 Cav3.1 단백질 발현 억제하는 시스템을 meidal septum에서 구현하여 쥐의 물체 탐색 능력을 증가시키는 기작을 재현하였다. 따라서, 본 논문에서는 광활성 Flp이 비침입성, 효율적인 특성과 쉽게 사용 가능한 광유전학 모듈로서 부작용 없고, 확장성 높은 유전자 발현 조절 기술로서 뇌과학 분야에서 널리 사용 가능한 기술임을 제시하고자 한다.
