5'-AZA-CdR (decitabine), known as a DNA-demethylation agent, is used as an anticancer agent for cancer such as colorectal cancer and acute myeloid leukemia. Recent studies have shown that decitabine abrogates the methylation of DNA regions that encoded endogenous retroviruses (ERVs) in cancer cells, resulting in an abnormal increase in intracellular ERV expression. Besides, these ERVs can make dsRNA structure form, leading to immune responses in cancer cells leading to apoptosis. However, unlike previous study results, some patients are not susceptible to chemotherapy. Moreover, dsRNAs that are abnormally increased by decitabine can interact with other dsRNA-binding proteins. Therefore there would be another critical factor that could exist. Through screening experiments to confirm the interaction effect between ERV and dsRNA binding proteins, we identified staufen1 as a protein that has a significant impact on the apoptotic mechanism of decitabine. Based on the results of this study, we expect to be able to realize patient-customized treatment with anticancer drugs.
탈 메틸화제로 알려진 데시타빈은 대장암과 혈액암 등에 항암제로 사용되고 있다. 최근 연구결과에 따르면 데시타빈이 암세포 내에서 내인성 레트로 바이러스(ERV)들이 암호화 되어있는 DNA 지역의 메틸화를 없앰으로써 세포 내 ERV 발현을 비정상적으로 증가시키고 ERV들이 double-stranded RNA(dsRNA) 구조를 이루면서 세포내 면역반응을 유도, 세포 사멸로 이끌어 낸다고 밝혀졌다. 하지만 현재까지의 연구결과와는 다르게 해당 항암제에 대한 감수성을 갖지 않는 환자들이 다수 있으며, 데시타빈에 의해 비정상적으로 증가한 dsRNA들은 또 다른 여러 dsRNA-binding protein들과 상호작용을 할 수 있기 때문에 해당 약물의 감수성을 설명할 수 있는 또 다른 요소들이 있을 것이라고 판단하였다. 스크리닝 실험을 통해 여러 dsRNA-binding protein들과 ERV 간의 상호작용에 의한 영향을 파악하였으며 큰 효과를 보인 staufen1이 작용하는 메커니즘을 연구하여 이를 해당 약물에 대한 감수성 예측 모델과 새로운 항암제 개발의 지표로서 활용하는데 가능성을 보았다. 본 연구의 결과를 바탕으로 항암제를 통한 환자 맞춤형 치료를 실현할 수 있는 밑거름이 될 것이라 기대한다.
