Cells are surrounded by other cells and interact with each other through close connection. This cell-cell adhesive interaction induces signaling pathways and affects the behavior of cells. Especially, in taste buds, gustatory cells and neuronal cells are closely located and information about taste is transmitted from tongue to brain through neuronal cells. In order to make gustatory and neuronal cells interact like taste buds, it is essential to bring both cells close together. In this study, I investigate DNA-PEG-DPPE polymers as a linker to closely connect gustatory cells and neuronal cells for mimicking actual taste buds with an aim to understand how gustatory and neuronal cells communicate with each other as a response to taste stimuli in taste buds. DNA-PEG-DPPE consists of 1,2-dipalmitoyl-glycerol-3-phosphatidylethanolamine, two thiol modified single stranded oligonucleotides: $(ACTG)_5$-SH and $(CAGT)_5$-SH, and succinimidyl-[(N-maleimidopropionamido)-tetracosaethyleneglycol] ester. The complementary single stranded oligonucleotides can bind to each other, and phospholipids can be introduced into the lipid bilayer of the cell membrane. Hippocampal neuronal cells from 18 days’ rat embryo and gustatory cells from 2 days’ neonatal mouse were seeded on a poly-D-lysine/laminin and collagen I coated culture slide, respectively. Each complementary DNA-PEG-DPPE was injected to the cultured neuronal cells and gustatory cells, and then the gustatory cells were simply placed on the neuronal cells for DNA-mediated cellular self-assembly. After 1~2 days, assembled neuronal and gustatory cells were treated by bitter taste stimuli and their response were recorded by monitoring the flux of intracellular calcium ions.
세포는 주변의 다른 세포와 물질들로 둘러 쌓여 있고 이들과 상호작용한다. 이러한 세포와 세포간의 밀접한 상호작용은 세포 간의 신호 전달을 유도하고 세포의 행동에 영향을 준다. 특히, 미각 기관인 혀의 미뢰에서는 미각세포와 신경세포가 가까이 붙어 상호작용하여 맛에 대한 정보를 뇌로 전달한다. 미뢰와 같이 미각세포와 신경세포가 상호작용하게 하기 위해서는 두 세포를 가까이 위치시켜야 한다. 본 연구에서는 DNA-인지질 중합체를 사용하여 미각세포와 신경세포를 가까이 연결하고, 두 세포가 맛 자극에 반응하여 일어나는 상호작용에 대해 연구하고자 한다. DNA-인지질 중합체는 상보적인 염기서열의 다른 DNA와 선택적으로 결합하는 DNA와 세포막의 주성분인 인지질 그리고 연결체 역할의 PEG로 구성되었다. 신경세포와 미각세포는 각각 태아 18일차 쥐의 해마체와 생후 2일 생쥐의 혀에서 추출하였다. DNA-인지질 중합체를 두 세포에 각각 처리하고 단순히 미각세포를 신경세포 위에 올리는 것으로 두 세포는 DNA-인지질에 의해 접합되었다. 접합한 두 세포에 쓴 맛 자극을 처리하고 세포의 반응을 세포 내 칼슘 농도 변화를 관찰하였다. 본 연구에서는 DNA-인지질에 의해 가까이 연결된 미각세포와 신경세포가 상호작용한 결과, 쓴 맛 자극에 대한 신호가 미각세포에서 신경세포로 전달되는 것을 확인할 수 있었다.
