Forster resonance energy transfer (FRET) based assay is an efficient method for the DNA detection in fast and simple way without complicated step. For practical application, DNA-functionalized CdTe/CdS core/shell quantum dots (QDs), which have long PL lifetime and high PLQY, are needed as an alternative donor instead of the organic dye. Synthesis of QDs is conducted in aqueous solution without ligand exchange and synthesized CdTe/CdS QDs show high PLQY of up to 77%. Furthermore, the PL lifetime of QDs is increased to several hundred nanoseconds with shell growth, which is induced by Type II band structure. DNA-functionalization is achieved directly during the shell growth of QDs. To increase the number of DNA on the surface of QDs and reduce the formation of CdS clusters, the ratio of core solution to shell precursor is optimized to 0.05:1. Since phosphorothioate modified DNA (ptDNA) exists on the surface of QDs, it can be confirmed that non-specific binding is prevented by the steric exclusion of ptDNA. As the result of investigation of target DNA detection using synthesized CdTe/CdS QDs, the target DNA can be detected up to about 13 nM concentration through steady-state PL analysis. In addition, CdTe/CdS QDs can solve the problem of the background signal from the direct excitation of acceptor. Because of its long PL lifetime, direct excitation of the acceptor can be excluded through time-resolved PL analysis. In this study, we synthesized ptDNA-functionalized water-dispersible CdTe/CdS QDs with high PLQY and applied its long PL lifetime for the first time to improve the sensitivity of FRET-based DNA detection.
형광 공명 에너지 전달 (Forster resonance energy transfer, FRET) 기반 분석법은 복잡한 단계없이 빠르고 간단하게 DNA를 검출할 수 있는 방법이다. 실제 DNA 감지에 적용하기 위해 유기염료를 대신하는 주개 물질로써 발광 효율이 높고 발광 수명이 긴 특성의 양자점이 필요하다. 이를 위해 리간드 치환없이 수용액 상에서 CdTe/CdS 코어쉘 양자점을 합성하였다. 합성된 양자점은 최대 77 %의 발광 양자 효율과 타입 II 밴드 구조에 따라 발광 수명이 수백 나노 초까지 긴 특성을 보인다. DNA 표면개질은 쉘 합성동안 이루어졌으며, 양자점 표면의 DNA 개수 증가와 CdS 클러스터 형성 억제를 위해 코어 용액과 쉘 전구체의 몰 비율은 0.05:1로 최적화하여 실험을 진행하였다. 양자점의 표면에는 phosphorothioate DNA (ptDNA)가 존재하기 때문에 입체 배제 효과에 의하여 비특이적 결합이 일어나지 않는 것을 확인할 수 있었다. 합성된 CdTe/CdS 양자점을 이용하여 FRET 기반 DNA 감지를 확인한 결과, 정상 상태 발광 분석을 통해 표적 DNA가 13 나노 몰까지 감지되는 것을 확인하였다. 또한 시간 분해 발광 분석을 통해 발광수명이 긴 양자점을 사용함으로써 기존의 한계였던 받개의 직접 여기 문제가 해결되는 것을 볼 수 있었다. 본 연구에서는 수용액 상에서 발광 양자 효율이 높고 발광 수명이 긴 ptDNA로 표면개질된 CdTe/CdS 양자점을 합성하여 FRET 기반 DNA 센서의 감도 향상을 위해 합성된 양자점의 발광 수명이 긴 특성을 처음으로 적용하였다.
