Axonal branches, one of the features of neuronal development, extending from one axon make it possible for individual neurons to establish synapses with multiple targets. This complex structural connection in a nervous system allows one neuron to transmit a wide range of signals to multiple neurons, which is essential for performing various functions of the nervous system such as learning, memory, sensation, and response. The Microtubules and the actin filament induced the formation of the axonal branches. In this work, we used an h-PDMS nanopillar array which was fabricated from AAO mold as a culture platform. We found that the h-PDMS nanopillar array not only promoted the growth of the axon, but also significantly increased the number of axonal branches of primary hippocampal neurons. Also, we found that h-PDMS nanopillar array induced condensation of F-actin on the axon shaft using various inhibitors. A study on the effect of topographical environment on axonal branch formation not only can contribute to expanding knowledge of the cell-biological mechanism of regulation of branch formation, but also contribute to the formation of the high-ordered neural system.
신경세포의 성장에 중요한 부분중 하나인 축색돌기 가지는 개별 세포가 여러 대상에 시냅스를 형성할 수 있도록 도와주는 중요한 역할을 한다. 신경세포에서 이러한 구조적 연결은 단일 세포가 다양한 세포에 신호를 전달할 수 있도록 도와주므로 학습, 기억, 감각, 반응과 같은 신경계에서 필수적이다. 주로 이러한 축색돌기 가지는 Microtubule과 actin filament에 의해 형성된다. 본 연구에서는 AAO 주형을 통해 만든 h-PDMS 나노기둥 기판을 사용하였다. 이 기판은 해마 신경세포의 길이성장 뿐만아니라 축색돌기 가지 형성에도 영향을 미친다는 것을 확인하였다. 또한 이 기판은 F-actin의 응축을 유도한다는 것을 다양한 억제제를 사용함으로써 확인하였다. 이 연구에서의 기판의 지형 환경이 축색돌기 가지형성에 미치는 영향은 세포 생물학에서의 가지 형성의 매커니즘 원리와 고차원적인 신경계를 이해하는데 기여를 할 것이라 생각한다.
