The numbers and types of cells constituting vertebrate neural tissues are determined by cellular mechanisms that couple neurogenesis to the proliferation of neural progenitor cells. Here, we identified a role of mammalian target of rapamycin complex 1 (mTORC1) in regulation of growth and development of mouse retina. We found that in mTORC1-hyperactive Tuberous sclerosis complex 1 (Tsc1)-deficient mouse retina, retinal progenitor cells (RPCs) undergo accelerated cell cycle progression and precocious neurogenesis. The evidences showed that mTORC1 activation facilitates both the synthesis and degradation of cyclin proteins to regulate mitotic cell cycle progression of RPCs and consequently the retinal neurogenesis. We showed that mTORC1 induces the expression of immunoproteasome component Psmb9 via signal transducer and activator of transcription 1 (Stat1) in the RPCs, and concomitant loss of Psmb9 decelerates cell cycle progression of RPCs in Tsc1-deficient mouse and normalizes retinal developmental schedule. Collectively, our results establish a developmental role for PI3K-Akt-mTORC1 pathway, showing that it promotes neural development by coupling synthesis and degradation of cell cycle regulator proteins in the progenitor cells through activation of protein turnover via a mechanism involving the immunoproteasome.
척추동물의 신경계는 발생과정에서 신경전구세포 (neural progenitor cell)가 분열(proliferation)을 반복하며 점차 신경세포와 교세포로 분화(differentiation)되면서 형성된다. 신경조직이 일정한 크기를 유지하고 정상적인 기능을 수행하기 위해선 이러한 신경전구세포의 분열 또는 분화과정이 균형을 이루어야 하며 이를 위해선 전구세포의 세포주기 진행의 안정적인 조절이 수반된다. 쥐 망막은 이러한 신경발생의 조절 과정의 메커니즘을 연구하는데 있어 매우 유용한 모델로써 망막을 구성하는 6종류의 신경과 1종류의 교세포는 모두 망막전구세포 (retinal progenitor cell)에서 분화되어 생성되지만 이 세포의 분열과 분화가 어떠한 기작으로 유지되고 조절되는지는 아직까지 명쾌히 밝혀지지 않았다. 본 논문에서는 mTORC1 신호전달계가 세포 내•외부적 신호에 대응해 세포 주기를 조절한다는 점에 착안, mTORC1의 활성이 세포주기 조절과 나아가 전구세포의 신경분화에 미치는 영향을 조사하였고, 생쥐의 망막 발생과정에서 mTORC1 신호전달계가 Immunoproteasome의 개입과 함께 신경 전구세포의 분열과 분화정도를 조절하는 요인임을 확인하였다. 생쥐 망막 전구세포내의 mTORC1 신호전달계가 과활성되면 정상 망막보다 전구세포의 세포주기 진행이 가속화되어 과분열이 일어나고 이와 함께 신경세포 분화 또한 증가하였으며 이는 mTORC1 신호전달계가 세포주기를 조절하는 Cyclin 단백질의 합성/분해 속도를 조절하면서 세포주기 진행을 가속화 시킨 영향임을 확인하였다. 흥미롭게도 mTORC1이 과활성된 망막에서 Immunoproteasome의 subunit 중 하나인 Psmb9 단백질이 정상보다 과발현되어 있었으며 이 단백질이 손실되었을 시 mTORC1 과활성의 영향으로 가속화된 전구세포의 세포주기 진행이 경감되는 것을 확인하였다. 이를 통해 결과적으로 mTORC1 pathway의 활성이 stat1 유전자를 매개로 하여 psmb9 의 발현을 유도, Immuno-proteasome의 개입과 함께 세포주기진행을 가속화시켜 나아가 신경발생과정에도 영향을 준다는 결론을 내렸다.
